花姜酮通过抑制NF-κB蛋白表达缓解重症急性胰腺炎大鼠肝损伤 被引量:5

Zerumbone on improving liver injury of rats with severe acute pancreatitis through inhibiting NF-κB protein expression

摘要 目的观察花姜酮对重症急性胰腺炎大鼠肝损伤的保护作用及对核因子-κB(NF-κB)蛋白表达的影响。方法将72只Wistar大鼠随机分为模型组、实验组及假手术组,每组24只。模型组及实验组于胰胆管逆行注射5%牛黄胆酸钠诱导构建重症急性胰腺炎大鼠模型,假手术组则注射等量0.9%NaCl。实验组大鼠分别于建模后6,12h时股静脉穿刺给予花姜酮溶液10mg·kg-1,模型组及假手术组则给予等量0.9%NaCl。用试剂盒检测大鼠血清肝功能指标水平,用免疫组化法检测肝组织NF-κB蛋白表达。结果建模后12h时假手术组、模型组及实验组血清天门冬氨酸转氨酶(AST)水平分别为(21.36±1.51),(463.39±101.31),(339.06±98.70)U·L-1;丙氨酸转氨酶(ALT)水平分别为(27.82±1.61),(199.81±61.23),(152.08±59.23)U·L-1;肝组织NF-κB蛋白免疫组化染色评分分别为(2.13±0.64),(10.04±0.71),(4.01±0.61)分。模型组分别与假手术组和实验组比较,差异均有统计学意义(均P<0.05)。结论花姜酮可通过抑制重症急性胰腺炎大鼠肝组织NF-κB蛋白表达发挥肝脏保护作用。 Objective To explore the protective effect of zerumbone on liver injury of rats with severe acute pancreatitis and influence on the nuclear factor-κB(NF-κB) protein expression. Methods A total of 72 Wistar rats were randomly assigned to model group, test group and sham-operation group, with 24 rats in each group. The model group and test group were injected with 5% sodium taurocholate in biliopancreatic duct to induce severe acute pancreatitis, sham-operation group was injected with 0.9%NaCl. Rats in test group were given 10 mg·kg-1 of zerumbone solution via femoral vein puncture at 6, 12 h after modeling, rats in model group and sham-operation group were given equal amount of 0.9%NaCl. The serum liver function parameters were measured by kits, and the expression of NF-κB protein in liver tissue was determined by immunohistochemistry. Results At 12 h after mode-ling, the serum aspartate aminotransferase(AST) levels in sham-operation group, model group and test group were(21.36±1.51),(463.39±101.31),(339.06±98.70) U·L-1, respectively;the alanine aminotransferase(ALT) levels were(27.82±1.61),(199.81±61.23),( 152. 08 ±59. 23) U·L-1,respectively;the immunohistochemistry stain scores of liver tissue NF-κB protein were2. 13 ± 0. 64,10. 04 ± 0. 71,4. 01 ± 0. 61,respectively. There were statistically significant differences between the model group and the sham-operation group/test group in the above indicators( P < 0. 05). Conclusion Zerumbone can play the role of liver protection through inhibiting the liver tissue NF-κB protein expression of rats with severe acute pancreatitis.
