摘要
目的:研究八宝哮咳散(BXS)的药效学及急性毒性。方法:选取50只昆明种小鼠随机分为对照组、蛤蚧定喘胶囊组及BXS高、中、低剂量组,采用小鼠氨水引咳法、气管段酚红法、蛋清性足肿胀法、胸腺和脾脏指数测定法,检测BXS镇咳、祛痰、抗炎作用、胸腺和脾脏指数。取50只英国种豚鼠随机分为对照组、蛤蚧定喘胶囊组及BXS高、中、低剂量组,采用组胺-乙酰胆碱引喘法检测BXS平喘作用。取40只昆明种小鼠随机分为对照组和BXS组,以最大给药剂量1日灌胃2次,给药后连续观察14 d,测定最大给药量(MFD)。结果:BXS高、中、低剂量组和蛤蚧定喘胶囊组小鼠咳嗽潜伏期均较对照组明显延长,小鼠咳嗽次数均较对照组显著减少,小鼠气管酚红排泌量、胸腺指数和脾脏指数均较对照组显著增加,小鼠足趾肿胀度均较对照组明显降低(均P<0.05)。BXS高、中、低剂量组和蛤蚧定喘胶囊组豚鼠哮喘潜伏期均较对照组明显延长(P<0.01)。急性毒性实验中,小鼠均无明显中毒反应,无死亡。小鼠MFD为生药144 g/kg。结论:BXS具有显著的镇咳、祛痰、抗炎、平喘作用以及改善免疫器官的作用,其小鼠MFD相当于成人日用量的320倍,未见明显毒性反应。
Objective: To research the effects of Babao Xiaokesan (BXS) on pharmacodynamic and acute toxicity. Methods: Fifty Kunming mice were randomly divided into control group, Gejie Dingchuan capsule group,and BXS high-, med- and low- dose groups. Using ammonia induced cough in mice, tracheal phenol red excretion, foot swelling method based on albumen and measurement of thymus index and spleen index to observe the effects of BXS on relieving cough, dispelling phlegm, anti-inflammation, thymus index and spleen index. Fifty British guinea pigs were randomly divided into control group, Gejie Dingchuan capsule group, BXS high-, med-, and low- dose groups. Using histamine-acetylcholine induced asthma in guinea pigs to observe the effect of BXS on relieving asthma. Forty Kunming mice were randomly divided into normal control group and BXS group, and the BXS group mice were treated with the maximum dose of BXS via gavage. After 14 days, the maximum feasible dose (MFD) was calculated. Results: Compared with the control group, the latent period of cough, phenol red excretion of tracheal, the thymus index and spleen index were increased, while the frequency of cough and the swelling degree of foot were decreased in BXS high-, med-, and low-dose groups and Gejie Dingchuan capsule group ( P <0.05). The latency of induced asthma of guinea pigs in BXS high, medium and low dose groups was significantly longer than that in the control group ( P <0.01). In the experiment of acute toxicity, there were no obvious toxic reactions and no case of death. The MFD given to the mice was 144 g/kg crude drug. Conclusion: BXS has significant antitussive, expectorant, anti-asthma, and anti-inflammatory effects and it can improve immune organs. The MFD of BXS in mice is 320 times the daily dosage of adult with no obvious toxic reaction.
