摘要
目的研究分析缝隙连接(Cx)蛋白及其mRNA在寻常型银屑病皮损中的表达及其与病情严重程度的相关性。方法选取2015年1月2017年12月我院接诊治疗的寻常型银屑病患者80例作为研究组,选取同期在我院进行体检的健康人80例作为对照组,采用银屑病皮损面积与严重程度指数(PASI)评分对患者病情进行评定,分别采用免疫组化和基因组扩增试剂盒检测Cx蛋白及其mRNA表达水平,比较研究组和对照组检测结果,对比2组间检测结果的意义,分析指标水平与病情严重程度的相关性。结果4组患者Cx26蛋白平均光密度值比较差异有统计学意义(P<0.05),研究组Cx26、Cx43蛋白及表达显著高于对照组(P<0.05);4组患者Cx26蛋白对应mRNA平均光密度值比较差异有统计学意义(P<0.05),各组Cx30、Cx31、Cx31.1、Cx32、Cx37及Cx45蛋白及其对应mRNA表达水平比较差异无统计学意义(P>0.05)。研究组Cx26、Cx43蛋白及其对应mRNA表达显著高于对照组(P<0.05),不同病情严重程度的患者Cx26和Cx43蛋白及其mRNA表达意义差异有统计学意义(P<0.05),Cx26(r=0.624,P<0.01)和Cx43(r=0.595,P<0.01)及其mRNA表达水平与PASI评分存在显著的正相关性。结论Cx26和Cx43及其mRNA在银屑病患者中的表达显著偏高,其表达水平与患者病情严重程度显著正相关,为银屑病的临床诊断和治疗中的病情评估有着一定的参考价值。
Objective In order to study and analyze the expression of connexin (Cx) and their mRNA in psoriasis vulgaris,as well as its correlation with the severity of psoriasis vulgaris.Methods Eighty patients with psoriasis vulgaris who received treatment in our hospital from January 2015 to December 2017 were selected as the study group,and 80 healthy people who taken medical examination in our hospital during the same period were selected as the control group.The patients' condition was assessed by psoriasis lesion area and severity index (PASI) score.Immunohistochemistry and genome amplification were used respectively.The expression level of Cx protein and its mRNA was detected by the kit.The results of the study group and the control group were compared.The differences of the results between the study group and the control group were compared.The correlation between the level of Cx protein and the severity of the disease was analyzed.Results The average optical density values of Cx26 protein in the four groups were significantly different (P<0.05).The expression of Cx26 and Cx43 protein in the study group was significantly higher than that in the control group (P<0.05).The average optical density values of Cx26 protein corresponding to mRNA in the four groups were significantly different (P<0.05).Cx30,Cx31.1,Cx32,Cx37 and Cx45 protein in each group were significantly different (P<0.05).There was no significant difference in the expression level of white and its corresponding genes (P>0.05).The expression of Cx26 and Cx43 protein and their corresponding mRNA in the study group was significantly higher than that in the control group (P<0.05).There were significant differences in the expression of Cx26 and Cx43 protein and their mRNA between patients with different severity of disease (P<0.05),Cx26 (r=0.624,P<0.01),Cx43 (r=0.595,P<0.01),Cx43 protein and their mRNA and PASI score were significant.The expression level of mRNA and PASI score had significant positive correlation.Conclusion The expression of Cx26,Cx43 and the
