摘要 目的:研究富马酸氯马斯汀(CLE)对心肌细胞缺血再灌注损伤的保护作用.方法:将培养的H9C2细胞通过缺氧4 h,复氧4 h,建立缺血再灌注损伤模型,实验随机分为3组:正常组、缺血再灌注组(MIRI组)、缺血再灌注加CLE后处理组(MIRI+CLE组).采用CCK-8法检测不同浓度CLE对缺血再灌注损伤心肌细胞活性的影响;比色法检测各组乳酸脱氢酶(LDH)、丙二醛(MDA)含量;透射电镜观察心肌细胞的超微结构;激光扫描共聚焦显微镜观察钙离子再分布状态.结果:CCK-8结果显示,培养基中CLE浓度为1.25×10^-3μg/μl时,H9C2细胞活力最强;比色法结果显示,MIRI组中LDH、MDA含量较正常组增高,MIRI+CLE组较MIRI组含量降低;透射电镜结果显示,与正常组相比,MIRI组细胞出现核膜模糊、线粒体空泡样变、髓样变,与MIRI组比较,MIRI+CLE组细胞的线粒体改变减轻;激光扫描共聚焦显微镜观察到,MIRI组细胞内钙离子荧光强度较MIRI+CLE组明显增强.结论:CLE减轻了缺血再灌注引起的H9C2心肌细胞损伤. Objective :To study the protective effect of clemastine fumarate on myocardial ischemia-reperfusion injury. Methods :Cultured H9C2 cells were randomly divided into 3 groups :normal group (CON group),ischemiareperfusion injury group( MIRI group),and ischemia-reperfusion injury plus clemasine fumarate post-treatment group (MIRI+CLE group). The effects of different concentrations of clemastine fumarate on myocardial cell activity in ischemia-reperfusion injury were detected by CCK-8 method. The content of LDH and MDA in each group was detected by colorimetry. The ultrastructure of cardiomyocytes was observed by transmission electron microscopy. Laser confocal microscopy was used to observe the redistribution state of calcium ions. Results :The results of CCK-8 showed that H9C2 cells had the strongest activity when the concentration of CLE in the medium was 1.25×10^-3 μg/μl. The colorimetric method showed that the content of LDH and MDA increased in MIRI group compared with CON group. the content of MIRI+CLE group decreased compared with MIRI group. Transmission electron microscopy showed that nuclear membrane nucleus and mitochondrial vacuolization were observed in MIRI group compared with CON group. Compared with MIRI group,mitochondrial changes in MIRI+CLE group were not obvious. Laser confocal microscopy showed increased intracellular calcium fluorescence intensity in MIRI group compared with MIRI+CLE group. Conclusion :Clemastine fumarate alleviates H9C2 cardiomyocyte injury induced by ischemia-reperfusion.
