基于TCGA数据库分析NMB基因在结肠腺癌的诊断和预后价值 被引量:2

摘要 目的探讨分析NMB基因在结肠腺癌的诊断和预后价值。方法从TCGA公共数据库下载NMB基因在结肠腺癌及癌旁正常组织中的mRNA表达量和对应病例的临床资料,然后绘制小提琴图比较NMB基因在结肠腺癌组织和癌旁正常组织的差异表达,绘制诊断性ROC曲线探讨其在结肠腺癌的诊断价值,另外运用Kaplan-Meier生存曲线、单因素和多因素Cox模型来探讨NMB基因的预后价值。结果小提琴图结果提示NMB基因在结肠腺癌癌组织的表达水平高于癌旁正常组织(P <0.001),诊断性ROC曲线结果说明NMB基因在结肠腺癌的诊断中有较高价值(P <0.0001, AUC=0.976,95%CI:0.963~0.990)。探索NMB基因的预后价值发现NMB与结肠腺癌患者总生存时间有关(Adjusted P=0.007,HR=1.778,95%CI:1.167~2.710),NMB低表达的结肠腺癌患者有更好的预后。结论 NMB基因可能是结直肠癌诊断和预后的一个独立潜在的生物学标志物。 Objectives To investigate the diagnostic and prognostic value of NMB gene in colon adenocarcinoma(COAD).Methods The expression of NMB mRNA in COAD tissue and paracancerous normal tissue, as well as respective patients’ clinical data were downloaded from TCGA public database. Violin plot was used to compare the expression of NMB gene in COAD tissue with that in paracancerous normal tissue. Receiver operating characteristic(ROC) curve was used to investigate the diagnostic value of NMB gene in COAD. Kaplan-Meier, univariate and multivariate COX regression were used to investigate the prognostic value of NMB gene. Results Violin plot showed that the expression of NMB gene was significantly higher in COAD tissue than in paracancerous normal tissue(P < 0.001). ROC curve showed that NMB gene had a significant diagnostic value for COAD(P < 0.001, AUC = 0.976, 95% CI: 0.963~0.990). NMB gene was significantly associated with overall survival(adjusted P =0.007, HR = 1.778, 95%CI: 1.167~2.710). Patients with lower NMB expression had better prognosis. Conclusion NMB gene could be a potential diagnostic and prognostic biological marker for COAD.
