摘要 目的探讨AGR2对低氧环境下肺癌细胞系A549迁移和侵袭的影响。方法低氧和常氧条件下培养A549细胞,通过Real-timePCR和Western blot法检测细胞中AGR2niRNA和蛋白的表达水平;低氧刺激A549的同时,使用AGR2特异性siRNA抑制其表达,通过平板划线实验和Transwell侵袭实验检测细胞迁移和侵袭能力;采用Kaplan-Meier Plotter分析AGR2差异表达对肺腺癌患者预后的影响。结果低氧微环境可显著上调A549细胞中AGR2的表达;敲低A549细胞中AGR2的表达,可显著抑制低氧介导的肺腺癌细胞迁移与侵袭;高表达AGR2的肺腺癌患者预后不良。结论AGR2参与低氧微环境下肺腺癌细胞系A549的迁移和侵袭转移。 Objective To investigate the effect of AGR2 on the migration and invasion of A549 cells under hypoxic microenvironment. Methods The AGR2 cells were cultured under normal and hypoxic condition. The expression levels of AGR2 mRNA and protein were detected by real-time PCR and Western blot, respectively. After stimulation by hypoxia and transfection with AGR2 siRNA, Wound healing assay and Transwell invasion assay were used to detect the migratory and invasive ability of A549 cells, respectively. Kaplan-Meier Plotter was used to analyze the effect of differential expression of AGR2 on the prognosis of patients with lung adenocarcinoma. Results The expression of AGR2 in A549 cells was significantly upregulated by hypoxia. Hypoxia-mediated migration and invasion of A549 cells were inhibited markedly by AGR2 knockdown. Lung adenocarcinoma patients with higher AGR2 expression had poor prognosis. Conclusion AGR2 is involved in migration and invasion of lung adenocarcinoma cell line A549 under hypoxic microenvironment.
