摘要
研究复方中药对热应激肉仔鸡生产性能、肝脏HSP70 m RNA水平与蛋白表达的影响。试验选取120只18日龄AA+肉仔鸡,随机分为4组进行试验,即复方中药组、维生素组、纯中药组和阳性对照组。试验结束,每组随机迅速宰杀试验鸡3只,采取肝脏组织,采用实时荧光定量PCR法、H.E染色和Western blot法对热应激肉仔鸡肝脏中HSP70 mRNA水平与蛋白表达进行测定。结果表明,复方中药组、纯中药组肉仔鸡平均日增重分别比阳性对照组增加10.87 g(P<0.01)和6.32 g(P<0.05),死淘率分别降低10%(P<0.05)和16.67%(P<0.01),而且复方中药组和纯中药组肉鸡肝脏中HSP70 mRNA水平显著提高,蛋白表达呈动态、不稳定变化。提示本复方中药可提高肉仔鸡机体细胞热耐受水平,提高生产性能。
To research on anti-chicken heat stress,regulation of HSP70 expression in liver and the effects of productivity by traditional Chinese medicine compound. 120 18-day old AA+ broilers were selected and randomly divided into four groups for the experiment,namely,traditional Chinese medicine compound group,vitamin group,pure traditional Chinese medicine group and high-temperature control group.3 experimental chickens were killed randomly and quickly in each group and the livers were collected. HSP70 mRNA and protein expression in liver of hot-stressed broilers were determined by Real-time quantitative PCR method,H.E staining and Western-blot method. The results showed that the average daily weight gain of broilers in the traditional Chinese medicine compound group and the pure Chinese medicine group increased by 10.87 g(P<0.01)and 6.32 g(P<0.05). The death rate was reduced by 10%(P<0.05)and 16.67%(P<0.01)respectively. The content of HSP70 mRNA in the liver of broilers in the traditional Chinese medicine compound group increased significantly. However,protein expression was dynamic and unstable.It were stated that the traditional Chinese medicine compound could improve the heat tolerance level of body cells and improve the production performance of broiler chickens.
