摘要
目的了解南京江宁区无偿献血者发热伴血小板减少综合征布尼亚病毒(SFTSV)感染情况。方法 2018年5-8月搜集来自献血者血清标本,运用全自动血液酶免筛查平台,采用双抗原夹心ELISA法检测血清中SFTSV总抗体(IgM和IgG)和IgM型抗体,并采用统计学方法对献血者的特点与感染相关性进行风险因素分析。采用荧光定量PCR方法检测抗体阳性标本的SFTSV RNA。结果本次调查的850份献血者血清标本中,SFTSV总抗体阳性血清8例,阳性率为0.94%;其中IgM阳性标本5例,阳性率为0.59%。分析献血者年龄、性别、职业与SFTSV感染相关性,发现性别、年龄与SFTSV的感染均没有统计学差异(P>0.05);献血者所从事的职业与SFTSV的感染具有显著的统计学差异(P<0.05),农民的SFTSV感染率最高,为4.55%。抗体阳性血清标本未检测到SFTSV RNA。结论本研究结果提示江宁地区献血者中存在较低的SFTSV感染率,无病毒血症。是否会通过血液传播,后续还需要扩大筛查区域和更多样本量进一步调查和分析。
Objective To explore the severe fever with thrombocytopenia syndrome bunyavirus(SFTSV) infection in the voluntary blood donors in Jiangning of Nanjing.Methods From May to August 2018,serum samples were collected from donors and screened total SFTSV antibodies(including IgG and IgM) and IgM on an automatic platform by double antigen sandwich ELISA kit.Repeat-reactive samples were tested SFTSV RNA using reverse transcription(RT)-real-time polymerase chain reaction(PCR) assay with Taqman probes.Results Eight(0.94%) samples were SFTSV antibodies positive and Five(0.59%) were IgM positive.The positive seroprevalence antibody rates in blood donors were compared by statistical methods between different groups and SFTSV infection.No correlation was found between different genders,ages and SFTSV infection.There was significant difference between different occupations and SFTSV infection.The positive rate in blood donors from hilly village was the highest(4.55%).No SFTSV RNA was found in SFTSV antibodies positive samples.Conclusion This study demonstrated that SFTSV infection existed in voluntary blood donors,but no viremia occurred.Whether or not SFTSV transmit through blood transmission also requires expanding screening areas and more sample sizes for further investigation and analysis.
