摘要
目的:建立HPLC法测定小儿感冒颗粒中绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸和连翘苷的含量。方法:AgilentC18色谱柱(4.6mm×250mm,5μm);流动相:乙腈-0.4%磷酸,梯度洗脱;检测波长:0~45min(327nm),45~83min(202nm);柱温:30℃;流速:1mL·min^-1,进样量10μL。结果:绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸、连翘苷浓度分别在0.01267~0.3801、0.1040~1.5600、0.00840~0.2526、0.03190~0.3190μg·mL^-1范围内呈良好的线性关系(r=0.9999,1.0000,0.9966,1.0000);平均加样回收率分别为97.89%、96.37%、96.63%和103.3%(n=6)。结论:所建立的方法可同时测定小儿感冒颗粒中的多种有效成分的含量。
Objective : To establish an HPLC method for determination of chlorogenic acid, luteolin, 3,5-O-dicaffeoyl quinic acid and forsythin in Xiao′erganmao Granules. Methods : The test was performed on Agilent C 18 (4.6 mm×200 mm, 5 μm) under the gradient elution of acetonitrile-0.4% phosphate. The column temperature was 30 ℃.The detection wavelengths were set as 327 nm for chlorogenic acid, luteolin, and 3,5-O-dicaffeoyl quinic acid;and 202 nm for forsythin. The flow rate was 1.0 mL·min^-1 . Results :The linear ranges of chlorogenic acid, luteolin, 3,5-O-dicaffeoyl quinic acid and forsyhin fell into 0.012 67-0.380 1 μg·mL^-1 ( r =0.999 9), 0.104 0-1.560 0 μg·mL^-1 ( r =1.000 0), 0.008 40-0.252 6 μg·mL^-1 ( r =0.996 6), and 0.031 90-0.3190 μg·mL^-1 ( r= 1.000 0). The average recoveries of chlorogenic acid, luteolin, 3,5-O-dicaffeoyl quinic acid and forsyhin ( n =6) were 97.89%, 96.37%, 96.63%, 103.3% respectively. Conclusion :The established method can be used for simultaneous determination of multiple components in Xiao′er Ganmao Granules.
