摘要
目的观察密骨胶囊对去卵巢大鼠骨内血管内皮生长因子(VEGF)及其受体表达的影响。方法取3月龄雌性Wistar大鼠,运用卵巢切除手术建立骨质疏松模型。模型成功后随机分为模型组、密骨胶囊组,同时设立对照组。密骨胶囊组予密骨胶囊混悬液3 m L/d,模型组和对照组予相同体积生理盐水。检测大鼠左侧股骨的骨密度;运用三点弯曲实验方法检测右侧股骨的生物力学性能;运用Real-time q PCR和Western blot检测股骨内VEGF、VEGF受体1(VEGFR1)及VEGF受体2(VEGFR2)的表达。结果模型组股骨骨密度低于对照组,密骨胶囊组股骨骨密度高于模型组,且均有统计学差异。卵巢切除后股骨生物力学性能指标降低,密骨胶囊可以改善股骨生物力学性能,但未见统计学差异。大鼠去卵巢后,骨内VEGF、VEGFR1和VEGFR2 mRNA和蛋白表达均降低,密骨胶囊可以促进骨内VEGF和VEGFR2 m RNA和蛋白表达,而VEGFR1 m RNA和蛋白表达未见明显变化。结论密骨胶囊可以增加骨质疏松大鼠的股骨骨密度,可能是通过促进骨内VEGF及其受体VEGFR2的表达实现的。
Objective To observe the effect of Migu Capsule on the expression of vascular endothelial growth factor(VEGF) and its receptor in ovariectomized rats. Methods Osteoporosis models were set up in 3-month-old rats through ovariectomy;the model rats were divided into 2 groups: model group and Migu Capsule group, and meanwhile a control group was set up with shams;suspension of Migu Capsule 3 m L/d was given to rats in Migu Capsule group while saline of same volume was given to rats in model group and control group;measurement of the bone mineral density(BMD) of left femur was made in the 3 groups;three-point bending test was applied in detecting the biomechanical properties of right femur while Real-time qPCR and Western blot method were applied in detecting the expressions of VEGF, VEGF receptor 1(VEGFR1) and VEGF receptor 2(VEGFR2). Results The femoral BMD in model group was lower than that in control group(P<0.05) while the femoral BMD in Migu Capsule group was higher than that in model group(P<0.05);Migu Capsule was of the effect to improve the decreased femur biomechanical properties due to ovariectomy while no statistical differences were observed;after ovariectomy, the expressions of VEGF, VEGFR1 and VEGFR2 decreased and Migu Capsule promoted the expressions of VEGF and VEGF2 while no obvious changes were found in the expression of VEGF1. Conclusions Migu Capsule can increase the femoral BMD in ovariectomized rats, probably by promoting the expressions of VEGF and its receptor VEGFR2 in bone.
