摘要
目的采用RNA干扰技术,研究人脑胶质瘤SHG44细胞对化疗敏感性的影响。方法用脂质体法转染人脑胶质瘤SHG44细胞株,通过RT-PCR检测癌胚抗原相关细胞黏附分子1(carcinoembryonic antigen-related cell adhesion molecule 1,CEACAM1)mRNA的变化,CCK-8法检测在化疗药物替莫唑胺(temozolomide,TMZ)作用下转染CEACAM1 siRNA后SHG44细胞增殖情况,分析凋亡情况。结果与正常对照组及阴性对照组比较,siRNA干扰组SHG44细胞CEACAM1基因mRNA表达受到抑制(P<0.01),经不同浓度TMZ作用后,siRNA干扰组SHG44细胞的增殖抑制明显被增强(P<0.01),而且SHG44细胞对TMZ的IC50降低(P<0.05),细胞凋亡率增加(P<0.01)。结论合成的针对CEACAM1基因的特异性siRNA能够抑制SHG44细胞中CEACAM1基因的表达,并能提高胶质瘤细胞的化疗敏感性。
Objective RNA interference technique was used to study the influence on chemosensitivity of human glioma SHG44 cells.Methods The siRNA specific for CEACAM1 was designed and synthesized,and transfected into human glioma SHG44 cells by lipofectamine.CEACAM1 m RNA expression was detected by RT-PCR.CCK-8 assay was used to detect the proliferation of SHG44 cells after transfection under the action of chemotherapy drug temozolomide(TMZ),and the flow cytometry was used to analyze cell apoptosis rate.Results Compared with those in control group and negative control group,the expression level of CEACAM1 gene m RNA in siRNA group was significantly decreased(P<0.01).After being treated with different concentrations of TMZ,the proliferation inhibition of SHG44 cells in the siRNA interference group was significantly enhanced(P<0.01).The IC50 to TMZ was significantly lower than that of control group(P<0.05),and apoptotic rate in siRNA group increased than that in other groups(P<0.01).Conclusion The siRNA specific for CEACAM1 can inhibit the expression of CEACAM1 gene in SHG44 cells and improve the chemosensitivity of glioma cells.
