摘要
目的观察肾衰宁胶囊对糖尿病肾病大鼠肾功能及Wnt/β-catenin信号通路的影响,探讨肾衰宁胶囊的肾保护作用。方法将50只健康SD大鼠随机分为正常组、模型组、肾衰宁低剂量组、肾衰宁中剂量组和肾衰宁高剂量组,每组10只。对除正常组外,其他组均进行糖尿病肾病造模。造模成功后,肾衰宁低、中、高剂量组分别灌胃0.11 g/kg、0.22 g/kg、0.44 g/kg肾衰宁胶囊,正常组、模型组灌胃等量生理盐水,均连续8周。末次给药24 h后,取各组大鼠血、尿,用于检测血肌酐(SCr)、尿素氮(BUN)及尿蛋白;取各组大鼠肾脏,分别进行HE染色、Masson染色,并采用Western-blot法检测肾组织中Wnt-1及β-catenin的相对表达量。结果模型组尿蛋白、SCr及BUN水平均明显高于正常组(P均<0.05),肾衰宁各剂量组尿蛋白、SCr及BUN水平均明显低于模型组(P均<0.05),且肾衰宁各剂量组呈剂量依赖性降低(P均<0.05)。HE染色显示肾衰宁各剂量组大鼠肾组织间质炎性浸润等病理改变明显轻于模型组,Masson染色显示肾衰宁各剂量组大鼠肾组织胶原沉积量明显少于模型组,且肾衰宁高剂量组病理改变更轻。模型组大鼠肾组织中Wnt-1及β-catenin相对表达量均明显高于正常组(P均<0.05),肾衰宁各剂量组大鼠肾组织中Wnt-1及β-catenin相对表达量均明显低于模型组(P均<0.05),且肾衰宁各剂量组呈剂量依赖性降低(P均<0.05)。结论肾衰宁胶囊可有效改善糖尿病肾病肾损伤,可能与其有效调节Wnt/β-catenin信号通路有关。
Objective It is to observe the effect of Shenshuaining capsule on renal function and Wnt/β-catenin signaling pathway in rats with diabetic nephropathy,and to explore its renal protective effect. Methods Fifty healthy SD rats were randomly divided into normal group,model group,Shenshuining low-dose group,Shenshuaining middle-dose group and Shenshenning high-dose group,with 10 rats in each group. Except for the normal group,the other groups were modeled for diabetic nephropathy. After successful modeling,the low-,medium-,and high-dose groups of Shenshuai Ning were intragastrically administered with 0. 11 g/kg,0. 22 g/kg,and 0. 44 g/kg Shenshenning capsules. The normal group and the model group were given the same amount of normal saline. All the rats were treated for 8 weeks. After 24 hours of the last administration,the blood and urine of the rats in each group were taken to detect serum creatinine( SCr),urea nitrogen( BUN) and urine protein. The kidneys of each group were taken for HE staining and Masson staining. The relative expression levels of Wnt-1 and β-catenin in renal tissues were detected by Western-blot method. Results The urine protein,SCr and BUN levels in the model group were significantly higher than those in the normal group( P < 0. 05). The urine protein,SCr and BUN levels in every Shenshuaining group were significantly lower than those in the model group( P < 0. 05),and the decrease of these levels was dependent on the dose in every group( P < 0. 05). HE staining showed that the pathological changes of renal interstitial inflammatory infiltration were not so serious in the rats in every Shenshenning group as that in the model group. Masson staining showed that the amount of collagen deposition in the kidney tissue of the rats in every Shenshuaining group was significantly less than that of the model group. The pathological changes in the high-dose group of Shenshuining were lighter. The relative expressions of Wnt-1 and β-catenin in the renal tissue of the model group were significantly higher
