摘要
目的观察芹菜素对腺嘌呤诱导的慢性肾衰竭(CRF)大鼠肾功能、血流动力学及血脂的影响,探讨其对CRF的保护作用。方法将100只SD大鼠按照随机数字表法随机分为正常组、模型组、芹菜素低剂量组、芹菜素中剂量组、芹菜素高剂量组,每组20只。采用腺嘌呤连续灌胃21d的方法复制CRF模型大鼠,芹菜素各剂量组腹腔注射给药治疗(1次/d),正常组和模型组同步给予生理盐水,疗程28d。称量体质量、肾脏质量并计算肾脏指数,经腹主动脉取血并测定肾功能监测指标,HE染色法对肾脏组织进行病理学检查并进行组织病变评分;测定肾脏组织血流量(RBF),测量尾动脉收缩压(SBP)、舒张压(DBP)和肾静脉压(RVP);测定血浆血脂指标。结果模型组大鼠体质量明显低于正常组(P<0.05),左侧肾脏质量、肾脏指数及明显高于正常组(P均<0.05);芹菜素中剂量组大鼠左侧肾脏质量、肾脏指数均明显低于模型组(P均<0.05);芹菜素高剂量组大鼠体质量明显高于模型组和芹菜素低剂量组(P均<0.05),左侧肾脏质量、肾脏指数均明显低于模型组和芹菜素低剂量组(P均<0.05)。模型组大鼠血清尿素氮(BUN)、血肌酐(SCr)、尿酸(UA)和24h尿蛋白量(Pro)、肾脏组织病变评分均明显高于正常组(P均<0.05);芹菜素中、高剂量组大鼠血清BUN、SCr、UA水平和Pro、肾脏组织病变评分均明显低于模型组(P均<0.05),且芹菜素高剂量组各指标均明显低于芹菜素低剂量组(P均<0.05)。模型组大鼠尾动脉SBP、DBP及血浆三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平均明显高于正常组(P均<0.05),RVP、RBF及血浆高密度脂蛋白胆固醇(HDL-C)水平均明显低于正常组(P均<0.05);芹菜素中、高剂量组大鼠尾动脉SBP、DBP及血浆TC、TG、LDL-C水平均明显低于模型组(P均<0.05),RVP、RBF及血浆HDL-C水平均明显高于模型组(P均<0.05);芹菜素高剂量组SBP、DBP及血浆TC、TG�
Objective It is to observe the effects of apigenin on renal function, hemodynamics and blood lipids in rats with adenine-induced chronic renal failure (CRF), and to explore its protective effect on CRF. Methods One hundred SD rats were randomly divided into normal group, model group, apigenin low dose group, apigenin middle dose group and apigenin high dose group according to random number table, 20 rats in each group. CRF model rats were replicated by continuous gavage of adenine for 21 days. The high, medium and low dose of apigenin groups were respectively given apigenin by intraperitoneal injection (1 time/d). The normal group and the model group were treated with the same volume of normal saline for 28 days. The body mass and kidney weight were weighed and the kidney index was calculated. The blood was taken from the abdominal aorta and the renal function monitoring indexes were measured. The pathological examination of the kidney tissue was performed by HE staining and the tissue lesion score was determined. The renal tissue blood flow (RBF), systolic blood pressure (SBP), diastolic blood pressure (DBP) of tail vein, renal venous pressure (RVP) and plasma lipids were detected. Results Compared with the model group, the treatment with medium and high dose of apigenin for 28 days can effectively improve the body weight of CRF rats, reduce kidney weight and reduce kidney index, and reduce renal function monitoring indicators: urea nitrogen (BUN), serum creatinine (SCr), uric acid (UA) and 24 h urine protein (Pro), significantly improved renal tissue lesions, reduced lesion scores, decreased SBP, DBP and increased RVP, RBF;significantly improved blood lipid monitoring indicators: reduced the contents of triglycerides (TG), total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C);the above differences were statistically significant ( P <0.05). Conclusion Apigenin has a pharmacological effect such as improving renal function and renal hemodynamics in CRF
