摘要
为了研究越橘菌根真菌的多样性,从吉林农业大学浆果园采集矮丛越橘(Vaccinium angustifolium)"美登"的根系,采用酸性品红乳酸甘油染色法观察菌根类型,用根段直接培养法分离根系共生真菌,利用真菌通用引物ITS1/ITS4扩增菌株的r DNA ITS序列; PCR产物测序后在Gen Bank中进行BLAST比对,然后采用非加权组平均法(UPGMA)进行聚类分析。结果表明:"美登"菌根类型包含典型的杜鹃花类菌根;试验共分离获得49株共生真菌,依据形态特征分为13个类型; ITS序列分析获得树粉孢属、镰孢属、膜盘菌属等9个属;聚类结果将其分为锤舌菌纲、粪壳菌纲和座囊菌纲3类。矮丛越橘"美登"的根系共生真菌物种丰富,优势或典型的共生真菌为杜鹃花类菌根(ERM)真菌--树粉孢属(Oidiodendron)真菌。
In order to study the diversity of blueberry mycorrhizal fungi,we collected the roots of lowbush blueberry(Vaccinium angustifolium) "Blomidon"from Jilin Agricultural University berry orchard. We used acid fuchsin staining method to observe the mycorrhizal morphology,applied root segments to culture fungi,and utilized the eukaryotic universal primer pair ITS1/ITS4 to amplify r DNA ITS sequences of fungus strains,then compared them with sequences in GenBank through a BLAST research before estimating them with UPGMA. The results show that the main infected mycorrhiza types of "Blomidon"is ericoid mycorrhiza. A total of 49 symbiotic fungi were isolated and classified into 13 types according to morphological characteristics. The ITS analyses show that the symbiotic fungi belong to 9 genera,including Oidiodendron,Fusarium,Hymenoscyphus,and so on.The phylogenetic tree divided them into 3 groups: Leotiomycetes,Sordariomycetes and Dothideomycetes. The conclusion is that the symbiotic fungi isolated from roots of lowbush blueberry "Blomidon"have rich species. Specially,Oidiodendron,a species of typical or dominant ericoid mycorrhizal fungi,has been found in this study.
