摘要
目的:研究薤白皂苷对冠心病寒痰阻滞证患者血小板聚集率及表面受体的影响,初步分析薤白皂苷的作用机制。方法:取冠心病寒痰阻滞证患者静脉血分离富含血小板血浆,按实验分组分别加入薤白皂苷等不同浓度受试药物,计算血小板聚集抑制率,流式细胞术检测ADP诱导活化血小板P-选择素(CD62P)、GPIIb-IIIa表达情况及对P2Y1和P2Y12受体的作用。结果:冠心病寒痰阻滞证薤白皂苷中、高浓度抑制ADP诱导的血小板聚集显著高于生理盐水对照的血小板聚集;4μmol/L的薤白皂苷显著降低活化血小板CD62p的表达率;4μmol/L的薤白皂苷下GPIIb/IIIa的表达率低于活化后表达率;低、中、高浓度的薤白皂苷分别联合P2Y1、P2Y12括抗剂对ADP诱导的血小板聚集率均显著高于ADP诱导的血小板聚集率。结论:薤白皂苷对于冠心病寒痰阻滞证患者ADP诱导的血小板聚集有更好的抑制作用,其可能作用于P2Y1和P2Y12,通过降低胞浆内巧离子浓度和升高CAMP含量抑制血小板聚集的效果。
Objective: To study the effects of Furostanol Saponins from Allium Macrostemon Bunge Bulbs( FS-AMBB)on Aggregation and Surface glycoprotein of platelet in patient with Coronary Heart Disease of Syndrome of blocking with cold and phlegm,Preliminary screened of the signal pathways by which FS-AMBB acts. Methods: Venous blood of patient with Coronary Heart Disease of Syndrome of blocking with cold and phlegm was screened and collected and platelets were isolated from venous blood. Concentrated platelets were divided into experimental groups with different concentrations of saponins and other test drugs to calculate the inhibition rate of platelet aggregation. Flow cytometry was used to detect ADP-induced activated platelet P-selectin( CD62 P),GPIIb-IIIa expression and P2 Y1 and P2 Y12 Receptor ’s role. Results:Inhibitory aggregation rate of platelets in patients with Syndrome of blocking with cold and phlegm with coronary heart disease was significantly higher than that of saline control group. 4 μmol/L of saponins significantly reduced the expression of activated CD62 p. There was no significant difference in the expression of GPIIb/IIIa between 4 μmol/L of saponins and GPIIb/IIIa of activated platelets. Low,medium and high concentrations of saponins and P2 Y1,P2 Y12 antagonists on ADP-induced platelet aggregation were significantly higher than ADP-induced platelet aggregation,and in a concentrationdependent manner. Conclusion: FS-AMBB can inhibit the aggregation of platelets,which are induced by ADP,in patients with Coronary Heart Disease of Syndrome of blocking with cold and phlegm and it may act on P2 Y1 and P2 Y12 to inhibit platelet aggregation in the way of concentrating Concentration and elevating the CAMP content in platelets.
