摘要
目的:观察芪蓟肾康汤含药血清对人肾小球系膜细胞的增殖及胞外信号调节激酶(ERK)通路的影响,探讨芪蓟肾康汤延缓肾小球硬化的作用机制。方法:采用血管紧张素Ⅱ(AngⅡ)诱导人肾小球系膜细胞增殖,采用血清药理学方法制备芪蓟肾康汤和替米沙坦含药血清。将人肾小球系膜细胞分为正常组、模型组、西药组、中药低剂量组、中药中剂量组和中药高剂量组六组,培养24、48、72 h后用4-甲偶氮唑蓝(MTT)法检测细胞增殖;培养48、72 h后用ELISA法检测的ERK1/2、pERK1/2含量。结果:与正常组相比,AngⅡ组能显著促进系膜细胞增殖,替米沙坦组和各浓度中药含药血清组均可不同程度抑制AngⅡ引起的系膜细胞增殖。AngⅡ刺激后系膜细胞中ERK1/2蛋白表达明显降低,pERK1/2蛋白的表达明显增高,替米沙坦和不同浓度中药含药血清干预后,ERK1/2蛋白水平显著升高、pERK1/2蛋白水平显著下降,其中中药高剂量组最为显著。结论:芪蓟肾汤可能通过抑制人肾小球系膜细胞增殖和ERK通路的磷酸化,从而达到延缓肾小球硬化的目的。
Objective:To observe the effect of Qiyi Shenkang Decoction on the expression of mesangial cell proliferation and the extracellular signal regulating kinase(ERK) pathway in human glomerular mesangial cells induced by AngⅡ. To explore the mechanism of Qiyi Shenkang Decoction in the treatment of glomerulosclerosis.Methods:The human glomerular membrane cells were divided into six groups: normal group, model group, Western medicine group, Qiyi Shenkang Decoction low, medium and high dose groups. After cultured for 24 h, 48 h, 72 h, cell proliferation was detected by 4-methyroazolam(MTT) method. The contents of ERK1/2 and pERK1/2 were detected by ELISA method after cultured for 48 h and 72 h.Results:Compared with the normal group, Ang Ⅱ group significantly promoted the proliferation of mesenchymal cells. Both the Timishatan group and Qiyi Shenkang Decoction groups could inhibit the proliferation of mesenchymal cells caused by Ang Ⅱ to varying degrees. The expression of ERK1/2 protein in the mesangial cells after AngⅡ stimulation was significantly reduced and the expression of TERK1/2 protein was significantly increased, and the levels of ERK1/2 protein and TERK1/2 protein decreased significantly after intervention with Timishatan and different concentrations of Qiyi Shenkang Decoction. Among them, the high dose group had the most significant effect.Conclusion:Qiyi Shenkang Decoction may delay glomerular sclerosis by inhibiting the proliferation of human glomerular membrane cells and phosphorylation of ERK pathway.
