摘要
目的研究E3泛素连接酶SIAH2与P-ERK在卵巢癌癌变过程中的表达及其与临床病理特征的关系。方法采用免疫组织化学S-P法检测159例卵巢石蜡包埋组织(60例卵巢良性囊肿、45例交界性肿瘤和54例卵巢上皮性癌)中SIAH2与P-ERK蛋白的表达情况,采用Western blot检测35例卵巢癌及癌旁卵巢组织中SIAH2与P-ERK的表达情况。结果免疫组织化学结果显示,在卵巢癌癌变过程中SIAH2与P-ERK蛋白表达阳性率呈递增趋势;统计学分析发现54例卵巢癌组织中SIAH2与P-ERK表达呈正相关(r=0.469,P=0.000);SIAH2与P-ERK与患者年龄及卵巢癌组织学类型无相关性(均P>0.05),与组织学分级和临床分期及淋巴结转移相关(P<0.05)。Western blot结果显示,卵巢癌中SIAH2与P-ERK蛋白的表达明显高于癌旁正常组织(P<0.05)。结论IAH2与P-ERK蛋白在卵巢癌癌变过程中发挥重要的作用,并且存在协调关系。
Objective To research the expressions of SIAH2 and P-ERK in carcinogenesis of ovarian cancer and the relationships with clinicopathological characteristics. Methods Immunohistochemical SP method was used to detect the expressions of SIAH2 protein and PERK protein in 159 paraffin-embeded ovarian tissue specimens( 60 cases of benign ovarian cyst,45 cases of borderline ovarian tumor,and 54 cases of epithelial ovarian cancer). Western blot was used to detect the expressions of SIAH2 and P-ERK in ovarian cancer tissue and ovarian tissue adjacent to ovarian cancer in 35 cases. Results The positive expression rates of SIAH2 protein and P-ERK protein in carcinogenesis of ovarian cancer showed an increasing trend. In 54 cases with ovarian cancer,SIAH2 expression was positively correlated with P-ERK expression( r = 0. 469,P = 0. 000). SIAH2 and P-ERK were not correlated with age and histological types( P>0. 05). SIAH2 and P-ERK were correlated with histological grade,clinical stage,and lymph node metastasis( P<0. 05). Western blot showed that the expression levels of SIAH2 protein and P-ERK protein in ovarian cancer tissue were statistically significantly higher than those in ovarian tissue adjacent to ovarian cancer( P<0. 05). Conclusion SIAH2 protein and P-ERK protein play important roles in carcinogenesis of ovarian cancer,and there is a coordination between the two indexes.
