摘要
目的分析长链非编码(LncRNA-TCL6)在胎盘植入组织中的表达水平及对滋养层细胞活性和迁移的影响。方法取2016年10月至2018年12月于我院行剖宫产合并胎盘植入的组织共40例,取40例正常胎盘组织作为对照,对比两组胎盘组织LncRNA-TCL6的表达水平,对HTR-8细胞采用RNA干扰敲低LncRNA-TCL6的表达水平后,采用CCK8检测其细胞活力情况,采用Transwell迁移实验和划痕实验评估细胞的迁移能力,采用Western-Blot 检测相关蛋白的表达水平。结果植入组的胎盘的LncRNA-TCL6 的相对表达量显著高于正常组(P<0.05),转染敲减HTR-8/Svneo细胞LncRNA-TCL6表达水平后,其细胞活性水平显著降低(P<0.05),且抑制HTR-8/Svneo细胞的迁移能力,敲减LncRNA-TCL6的HTR-8/Svneo细胞其E-Cadherin 表水平显著上升,而Vimentin、N-Cadherin的表达水平显著下降。结论LncRNA-TCL6 在胎盘植入组织中表达显著升高,且LncRNA-TCL6参与滋养层细胞的增殖和迁移作用。
Objective To analyze the expression level of LncRNA-TCL6 in placenta-implanted tissues and its effect on trophoblast cell activity and migration.Methods Forty patients with cesarean section and placental implatation in our hospital from October 2016 to December 2018 were enrolled as the implanted group and another 40 normal placental tissues as controls.The expression levels of LncRNA-TCL6 in the placenta were compared.After knocking down the expression level of LncRNA-TCL6 by HTR-8 cells,the cell viability was detected by CCK8.Transwell migration assay and scratch assay were used to assess cell migration ability,and the expression level of related proteins was detected by Western blot.Results The relative expression of LncRNA-TCL6 in the placenta of the implanted group was significantly higher than that in the control group(P < 0.05).After transfection and knockdown of HTR-8/Svneo cells LncRNA-TCL6 expression level,the cell activity level was significantly decreased(P < 0.05).Moreover,the migration ability of HTR-8/Svneo cells was inhibited,and the level of ECadherin was significantly increased in HTR-8/Svneo cells knocked down by LncRNA-TCL6,while the expression levels of Vimentin and N-Cadherin were significantly decreased.Conclusion LncRNA-TCL6 is significantly increased in placenta-implanted tissues,and LncRNA-TCL6 is involved in the proliferation and migration of trophoblast cells.
