AR和SKP2在三阴性乳腺癌中的表达及临床意义 被引量:5

Expression and Clinicopathologic Significance of AR and SKP2 in Triple-negative Breast Cancer

摘要 目的分析雄激素受体(androgen receptor,AR)、S期激酶相关蛋白2(S-phase kinase-associated protein2,SKP2)在三阴性乳腺癌(triple-negative breast cancer,TNBC)中的表达及它们与临床病理特征的关系,并对AR、SKP2在TNBC中的表达进行相关性分析。方法采用免疫组织化学(immunohistochemistry,IHC)Envision法检测96例TNBC和35例乳腺腺病/纤维腺瘤中AR、SKP2的表达,采用Westernblot法和RT-PCR法分别检测20例TNBC和5例癌旁乳腺组织中AR、SKP2的表达,分析它们与各临床病理特征的关系。结果IHC结果显示TNBC中AR的阳性表达率较乳腺良性病变低(P<0.05),SKP2在TNBC中的阳性表达率较乳腺良性病变显著升高(P<0.05);AR、SKP2在TNBC中的阳性率分别为18.8%、43.8%,AR与淋巴结转移状态显著相关(P<0.05),无淋巴结转移的肿瘤组AR的阳性表达率较高,AR与年龄、肿瘤直径、组织学级别、神经/脉管受累情况无相关性(P>0.05),SKP2与组织学级别、神经/脉管受累情况显著相关(P<0.05),组织学级别较高、神经/脉管受累的肿瘤组SKP2的阳性表达率较高,SKP2与年龄、肿瘤直径、淋巴结转移状态无相关性(P>0.05)。AR、SKP2在TNBC中表达显著相关(P<0.05)。RT-PCR结果显示在20例TNBC中AR、SKP2的mRNA高水平表达的组织均为8例,其中AR、SKP2共同高水平表达的组织有3例;5例癌旁乳腺组织有3例AR高水平表达,SKP2均低水平表达。同时Westernblot法检测显示相似的结果。结论AR在TNBC中显示较好的预后特征,SKP2在TNBC中显示较差的预后特征,提示它们在TNBC中是否能作为潜在的治疗靶点具有进一步的研究意义。 Objective To explore the expression of androgen receptor (AR) and s-phase kinase associated protein2 (SKP2) in triple-negative breast cancer (TNBC) and their relationship with clinical pathological features,and to analyze the correlation between the expression of AR and SKP2 in TNBC.Methods Envision method of Immunohistochemistry(IHC) was performed to detect the expression of AR and SKP2 in 96 cases of TNBC and 35 cases of breast adenosis and fibroadenoma,Western blot and real-time reverse transcription-polymerase chain reaction (RT-PCR) were applied to detect the expression of AR and SKP2 in 20 cases of TNBC and 5 cases of adjuvant cancer tissues,and the results were analyzed for their relationship with each clinical and pathological feature.Results IHC showed the expression rate of AR in TNBC was lower than that in breast adenosis and fibroadenoma ( P <0.05),and the expression rate of SKP2 in TNBC was significantly higher than that in breast adenosis and fibroadenoma ( P <0.05).The expression rate of AR and SKP2 in TNBC were 18.8%,43.8% respectively.AR was associated with a significant state of lymph node metastasis ( P <0.05).The expression rate of AR in the tumor group with no lymph node metastasis was higher comparatively,but there were no significant correlations between AR expression and age,tumor size,histological classification,neural/vascular involvement in TNBC ( P >0.05).There were significant correlations between SKP2 expression and histological classification,nerve/vascular involvement in TNBC ( P <0.05).The expression rate of SKP2 in the tumor group with higher histological classification or nerve/vascular invasion was higher,while there were no significant correlations between SKP2 expression and age,tumor size,lymph node metastasis in TNBC ( P >0.05).The expression of AR and SKP2 was significantly correlated in TNBC ( P <0.05).Eight cases in 20 TNBC showed higher mRNA expression of AR and SKP2.Higher coexpression of AR and SKP2 occurred in 3 cases.Three cases revealed higher AR while all cases in
