摘要
为培育芹菜雄性不育材料提供分子基础,对芹菜六合黄心芹(Q1)和津南实芹(Q2)线粒体atp6基因进行克隆,并对其花发育过程中基因的表达水平进行分析。结果表明:2种芹菜均能克隆到长短不同的2种atp6基因,其中长片段(Q1-L和Q2-L)atp6基因长度均为954bp,短片段(Q1-D和Q2-D)atp6基因长度为819bp。对芹菜atp6氨基酸进行分析,六合黄心芹和津南实芹长片段atp6基因(Q1-L和Q2-L)推导的氨基酸序列属于同一分枝,而较短片段atp6基因(Q1-D和Q2-D)推导的氨基酸进化上存在一定的距离。花发育过程中,2种芹菜长片段atp6基因相对短片段atp6基因表达量较高,表明长片段atp6基因可能与芹菜花发育相关。
To provide molecular basis for the cultivation of celery male sterile materials,the atp6 genes were cloned from the mitochondrial DNA of Liuhe Huangxin Qin(Q 1)and Jinnan Shiqin(Q 2),the evolution and the expression levels of the atp6 gene were analyzed during flowers development in the two celery cultivars.The results showed that two different long and short fragments of atp6 genes were existed in Liuhe Huangxin Qin and Jinnan Shiqin.The longer fragments(Q 1-L and Q 2-L)were 954 bp,while the shorter fragments(Q 1-D and Q 2-D)were 819 bp.Comparative analysis of the Neighbor Joining of phylogenetic analysis revealed that the longer atp6 genes(Q 1-L and Q 2-L)deduced amino acid sequences of two celery cultivars were clustered in same branch,while the short sequence atp6 genes deduced amino acid(Q 1-L and Q 2-L)of two celery cultivars have a little distance in the phylogenetic tree.The expression levels of the longer atp6 genes were higher than that of the shorter atp6 genes during flowers development in the two celery cultivars.Those results suggested that the long fragment of atp6 gene may be involved in flower development of celery.
