摘要
通过荧光光谱法研究了β-环糊精(β-CD)对溶菌酶(Lyz)与十六烷基三甲基溴化铵(CTAB)相互作用过程中Lyz构象的影响。结果表明,在25℃,pH=6.5和8.5的情况下,CTAB与Lyz摩尔比分别达到228和286时,Lyz仍可与CTAB在溶液中形成稳定的复合体,Lyz的构象不会发生明显的变化;在Lyz与CTAB形成的稳定复合体的溶液中加入β-CD,当β-CD与CTAB摩尔比达到1.2时,Lyz的荧光强度可以几乎完全恢复。因此,Lyz与CTAB的相互作用具有可逆性的特点。交联壳聚糖固载β-环糊精微球具有与单体β-CD类似的作用,可以代替传统人工伴侣系统中的剥离剂。
The effect ofβ-cyclodextrin(β-CD)on the lysozyme(Lyz)conformation during the interaction between Lyz and cetyltrimethylammonium bromide(CTAB)was studied by fluorescence spectroscopy.The results show that Lyz can still form a stable complex in solution with CTAB at 25℃,even when the molar ratio of CTAB to Lyz reaches 228 and 286 at pH=6.5 and 8.5 respectively,and the Lyz conformation remains unchanged significantly.The fluorescent intensity of Lyz is almost completely regained when the molar ratio ofβ-CD to CTAB reaches 1.2 by addingβ-CD to the stable complex solution of Lyz and CTAB.Therefore,the interaction between Lyz and CTAB has characteristic of reversibility.Cross-linked chitosan microsphere modified byβ-cyclodextrin would replace the stripping agent in the traditional artificial chaperone system,because it has a similar effect to that of the monomerβ-CD.
