侵袭性垂体泌乳素腺瘤中GPER1基因表达的变化

摘要 目的研究垂体泌乳素腺瘤中GPER1的表达及其在侵袭性肿瘤中基因表达的变化。方法选取2012—2017年复旦大学附属上海市第五人民医院神经外科收治的泌乳素垂体瘤患者,根据其头颅MR表现分为侵袭性组(n=37)和非侵袭性组(n=43),收集两组患者临床数据及手术切除的垂体瘤组织标本进行实验研究。采用免疫组化和荧光实时定量聚合酶链反应(RT-PCR)方法分别测定GPER1在侵袭性垂体泌乳素腺瘤中蛋白质及mRNA水平。结果两组患者的性别、年龄、术前血清泌乳素水平差异无统计学意义。侵袭组GPER1蛋白质水平低于非侵袭组[3.8(2.2~6.6)vs.11.2(6.0~12.0),P<0.001]且与性别、瘤体大小及术前血清泌乳素水平无关。RT-PCR结果显示侵袭组中GPER1 mRNA水平为0.47(0.03~2.3),非侵袭组为1.93(1.00~4.70),差异有统计学意义(P=0.021)。垂体泌乳素腺瘤中均有GPER1mRNA表达,但在侵袭组肿瘤中GPER1基因表达的水平较非侵袭性组低,两者之间差异有统计学意义。结论GPER1在垂体泌乳素腺瘤中起肿瘤抑制作用,且GPER1基因的低表达可能是引起泌乳素垂体瘤侵袭性增加的原因之一。 Objective To investigate the expression of GPER1 in invasive pituitary prolactin adenomas. Methods From 2012 to 2017,patients with prolactin pituitary tumor admitted by the Department of Neurosurgery,the Fifth People’s Hospital of Shanghai,Fudan University were divided into invasive group ( n =37) and non-invasive group ( n =43) according to their cranial MR manifestations.Clinical data and tissue samples of pituitary tumor resected by surgery of the two groups were collected.Immunohistochemistry and reverse transcription polymerase chain reaction (Rt-PCR) were used to observe the changes of protein and gene level of GPER1 in invasive pituitary prolactin adenomas. Results There was no difference in gender,age or preoperative prolactin level between the two groups.The protein expression of GPER1 in the invasive group was lower than that in the non-invasive group [3.8 (2.2-6.6) vs.11.2 (6.0-12.0),P <0.001],and was independent with gender,tumor size and preoperative prolactin level.Rt-PCR results showed that GPER1 mRNA expression was 0.47 (0.03-2.3) in the invasive group and 1.93 (1.00-4.70) in the non-invasive group ( P =0.021).The expression of GPER1 gene was found in all pituitary prolactin adenomas,but the GPER1 mRNA level in the invasive group was significantly lower than the non-invasive group ( P < 0.05). Conclusions The low expression of GPER1 gene may be related to the aggressive behavior of prolactin pituitary tumors.
