摘要
以海洋来源的芽孢杆菌(Bacillussp.)LYH18为出发菌株,优化其发酵条件,并对其所产肌氨酸氧化酶(SOX)进行分离纯化。结果表明,其最适产酶发酵条件为肌酸添加量1.0%,酵母膏添加量0.8%,MgSO4·7H2O添加量0.05%,KH2PO4添加量0.2%,K2HPO4添加量0.05%,初始pH值为7.0,培养温度25℃,装液量40mL/100mL。在此最佳发酵条件下,肌氨酸氧化酶酶活为4.45U/mL。通过十二烷基硫酸钠-聚丙酰胺凝胶电泳(SDS-PAGE)分析肌氨酸氧化酶,其纯化后的分子质量为43kDa。
Using Bacillus sp. LYH18 from ocean as the starting strain,the fermentation conditions were optimized,and the sarcosine oxidase (SOX) was isolated and purified. The results showed that the optimum fermentation conditions were creatine 1.0%,yeast extract 0.8%,MgSO4·7H2O 0.05%,KH2PO4 0.2%,K2HPO4 0.05%,initial pH 7.0,culture temperature 25 ℃,and loading volume 40 ml/100 ml. Under the optimum fermentation conditions,the activity of SOX was 4.45 U/ml. The molecular weight of the purified SOX was 43 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis.
