摘要
铜是生物体内不可缺少的金属元素,在机体的生物化学中起着至关重要的作用,人体内大多数铜离子都是与蛋白质相结合的。本文总结了铜离子及铜结合蛋白在癌症中的作用以及铜结合蛋白与癌症的关系,并讨论其增强铂类化疗药物的作用。我们特别关注铜结合蛋白如细胞运动介质1(MEMO1)、赖氨酰氧化酶(LOX)、LOX样蛋白和富含半胱氨酸的分泌蛋白(SPARC)如何从分子水平调节化疗药物。由于铜在癌症侵袭、迁移过程具有重要作用,故铜结合蛋白可能成为癌症防治的新靶点。
Copper is an indispensable metal ion in every organism. It plays a vital part in the process of biochemistry in each organism. Most of the copper ions in the body are protein-bound. This review summarized the roles the copper ions and copper-binding proteins play in cancer development, and the relationship between copper-binding protein and cancer. The roles were also discussed of copper-binding protein in enhancing efficiency of platinum-based chemotherapy. Especially, in this article, we also discussed how copper-binding proteins like mediator of ErbB2-driven cell motility 1 (MEMO1), lysyl oxidase (LOX), LOX-like proteins and secreted protein acidic and rich in cysteine (SPARC) regulate chemotherapeutic drugs at the molecular level therapy. Considering the importance of copper in the process of tumor invasion and metastasis, a further understanding of the function of copper-binding proteins may provide new targets for cancer control.
