摘要
目的:探讨血管生成素样蛋白1(ANGPTL1)在结直肠癌(CRC)中表达及对其生物学行为的影响。方法:采用免疫组织化学法检测ANGPTL1在CRC组织和癌旁正常组织中的表达,分析ANGPTL1表达与CRC患者临床病理特征的关系,并利用过表达的慢病毒转染HCT116细胞构建过表达ANGPTL1的HCT116细胞系,通过CCK8、Transwell小室实验研究过表达ANGPTL1对结直肠癌细胞增殖、迁移及侵袭等生物学行为的影响。结果:ANGPTL1在CRC组织阳性表达率分别为39.51%(32/81),明显低于癌旁正常组织中的80.25%(65/81),差异有统计学意义(P<0.05)。ANGPTL1表达与CRC患者性别、部位、组织学类型无关(P>0.05);与分化程度、Dukes分期、淋巴结转移有关(P<0.05);HCT116细胞增殖、迁移及侵袭能力略高于阴性对照组病毒原液(HCT116-control),但差异无统计学意义(P>0.05);与HCT116-control及HCT116细胞相比,ANGPTL1过表达的病毒原液(HCT116-ANGPTL1)增殖、迁移及侵袭能力显著提高(均P<0.01)。结论:ANGPTL1在结直肠癌中呈低表达,并与分化程度、Dukes分期、淋巴结转移相关,且过表达ANGPTL1后,CRC细胞迁移、增殖、侵袭能力明显提高。
Objective: To investigate the significance of angiopoietin-like protein 1(ANGPTL1) expression in colorectal cancer(CRC) tissues and its effect on biological behavior. Methods: The expression levels of ANGPTL1 in CRC tissues and adjacent normal mucosa were detected by immunohistochemistry, and its relationship with clinicopathological features of CRC patients was also analyzed. Meanwhile, the overexpression of HCT-ANGPTL1 cell line was established through virus transfection. Thereafter the effects of overexpression of ANGPTL1 on the proliferation, migration and invasion of colorectal cancer cells were studied by CCK8 and Transwell chamber experiments. Results: The positive expression rate of ANGPTL1 in CRC tissues was significantly lower than that in normal adjacent mucosa tissues[39.51%(32/81) vs 80.25%(65/81), P<0.05];Difference in the expression level of ANGPTL1 between CRC patients with different gender, lesion sites and histology was not statistic(P>0.05). The expression levels of ANGPTL1 in CRC patients with different degrees of differentiation, Dukes stage, and lymph node metastasis were statistically different(P<0.05). The proliferation, migration and invasion ability of HCT116-ANGPTL1 were significantly higher than those of HCT116 cell line and HCT116-control(P<0.01), while no statistic difference was found between HCT116 cell line and HCT116-control(P>0.05). Conclusion: ANGPTL1 is lowly expressed in colorectal cancer, which is associated with differentiation, Dukes stage, lymph node metastasis, and the overexpression of ANGPTL1 will result in the increase of CRC cell migration, proliferation, and invasion ability.
