摘要
目的水疱性口炎病毒纯化抗原制备兔抗VSV多克隆抗体,免疫学方法检测其结果分析比较。方法纯化的抗原制备兔抗VSV多克隆抗体,采用双向琼脂扩散试验,间接ELISA方法和中和试验测定效价。结果兔抗VSV多克隆抗体最高效价双向琼脂扩散试验为1∶32,间接ELISA法为1∶640,中和试验为1∶256。结论试验结果分析比较表明,建立纯化抗原制备的兔抗VS多克隆抗体,中和试验特异性好、敏感性高,可做为VSV的血清学检测和流行抗学调查抗供技术保障。
Objective To prepare rabbit anti-VSV polyclonal antibody by vesicular stomatitis virus purified antigen, and compare the results by immunological methods. Methods Rabbit anti-VSV polyclonal antibody was prepared by purified antigen. The titer was determined by two-way agar diffusion test, indirect ELISA method and neutralization test. Results The highest titer of the rabbit anti-VSV polyclonal antibody was 1∶32 for the two-dimensional agar diffusion test, 1∶640 for the indirect ELISA, and 1∶256 for the neutralization test. Conclusion The experimental results show that the rabbit anti-VSV polyclonal antibody prepared by the purified antigen has good specificity and high sensitivity, which can provide technical support for serological detection and epidemiological investigation of VSV.
