The expression of serum miRNAs in patients with myasthenia gravis and its clinical significance

摘要 目的探讨重症肌无力患者血清miRNAs的表达情况,为重症肌无力的早期诊断及治疗提供依据。方法 分析健康对照组(n=60)和重症肌无力组(n=60)的呼吸肌耐力、肺活量、最大通气量、肌无力水平评分等四项指标,进行统计分析;此外,收集健康对照组和重症肌无力组血清样本,进行实时定量聚合酶链式反应(qRT-PCR),检测血清中miRNAs的表达情况;收集重症肌无力患者治疗前后血清样本,进行qRT-PCR反应,检测血清中miRNAs的表达是否发生变化。结果 与健康志愿者相比,miR-107、miR-25、miR-20b在重症肌无力患者血清中表达降低,而miR-451、miR-29b和miR-27a表达升高;与治疗前相比,miR-107、miR-25、miR-20b在重症肌无力患者治疗后血清中表达呈升高趋势,miR-451和miR-27a无差异,miR-29b表达显著降低。结论 miR-107和miR-29b在重症肌无力患者中呈差异性表达,可能参与重症肌无力的发生和发展,可作为早期诊断及治疗重症肌无力的新靶点。 Objective To investigate the expression of miRNAs in the serum of patients with myasthenia gravis,and to provide evidence for early diagnosis and treatment of myasthenia gravis. Methods Four indexes including respiratory muscle endurance,lung capacity,maximum air capacity and muscle weakness level score were analyzed in healthy control group (n=60) and myasthenia gravis group (n=60).Serum samples from clinical healthy volunteers and patients with myasthenia gravis were collected for real-time quantitative polymerase chain reaction (qRT-PCR) to detect the expression of miRNAs in serum.Serum samples from patients with myasthenia gravis before and after treatment were collected for qRT-PCR analysis,which was used to detect whether the expression of miRNAs in serum was dysregulated. Results Compared with healthy volunteers,the expression of miR-107,miR-25,miR-20b was decreased in the serum of patients with myasthenia gravis.And while the expression of miR-451,miR-29b and miR-27a was increased.The expression of miR-107,miR-25 and miR-20b was increased in the serum of patients with myasthenia gravis after treatment.While,miR-451 and miR-27a were not changed,and miR-29b expression was significantly decreased compared with the expression before treatment. Conclusion miR-107 and miR-29b are differentially expressed in patients with myasthenia gravis ,which may be involved in the development and progression of myasthenia gravis.And they might be used as a new target for early diagnosis and treatment of myasthenia gravis.
