雄激素对SAMP8小鼠学习记忆能力的影响

摘要 目的:探讨雄激素对SAMP8小鼠学习记忆能力及海马CA1区胶质纤维酸性蛋白(GFAP)表达的影响。方法:21只7月龄雄性SAMP8小鼠随机分为假手术组、去势组和雄激素组,每组7只,7只同月龄SAMR1小鼠作为对照组。经去势及雄激素补充治疗后通过Morris水迷宫实验检测各组小鼠学习记忆能力,通过免疫组织化学实验检测海马CA1区GFAP的表达。结果:与其他3组比较,去势组1~5天5个时间点逃避潜伏期[(115.13±4.73)、(113.12±5.22)、(110.19±7.12)、(108.05±8.07)、(106.08±6.39)s]明显延长,跨越平台次数[(3.35±1.34)次]明显减少;与假手术组5个时间点[(98.87±13.37)、(93.22±13.36)、(88.01±7.82)、(83.71±11.12)、(78.13±10.06)s]、跨越平台次数[(6.96±1.40)次]比较差异有统计学意义(P<0.05);与去势组比较,雄激素组逃避潜伏期[(102.97±12.00)、(96.87±13.37)、(90.87±13.77)、(85.94±8.74)、(81.17±7.05)s]明显缩短,跨越平台次数[(7.43±1.75)次]明显增多,差异有统计学意义(P<0.05);去势组海马CA1区GFAP的表达明显增多,吸光度[(0.32±0.06)A]高于假手术组[(0.21±0.03)A](P<0.05),雄激素补充治疗后GFAP表达减少,吸光度[(0.22±0.05)A]低于与去势组(P<0.05)。结论:雄激素补充治疗可以改善小鼠学习记忆能力,可能通过减少GFAP表达,抑制星形胶质细胞活化实现。 Objective:To investigate the effects of androgen on learning and memory ability and GFAP expression in hippocampus of SAMP8. Methods: 21 seven-month-old male SAMP8 were randomly divided into sham operation group,castration group,and androgen group,with 7 mice in each group. 7 SAMR1 of the same age were used as control group. The learning and memory ability of mice in each group was detected through the Morris water maze test and the expression of GFAP in hippocampal CA1 region was detected and analyzed by immunohistochemical straining after castration and androgen supplementation therapy. Results: Compared with other three groups,the escape latency of the castrated group [(115.13±4.73),(113.12±5.22),(110.19±7.12),(108.05±8.07),(106.08±6.39) s] was significantly prolonged,the number of crossing the platform (3.35±1.34) was significantly reduced,and there was significant difference between the castrated group and sham operation group [(98.87±13.37),(93.22±13.36),(88.01±7.82),(83.71±11.12),(78.13±10.06) s],(6.96±1.40)( P <0.05). Compared with the castrated group,the escape latency of the androgen group [(102.97±12.00),(96.87±13.37),(90.87±13.77),(85.94±8.74),(81.17±7.05) s]was significantly shortened,the number of crossing platforms (7.43±1.75) was increased significantly,and the difference was statistically significant ( P <0.05). The expression of GFAP in hippocampal CA1 region of the castrated group was significantly increased,and the absorbance [(0.32±0.06) A]was higher than that of the sham operation group [(0.21±0.03) A]( P <0.05). The expression of GFAP was decreased after androgen replacement therapy,and the absorbance [(0.22±0.05) A] was lower than that of the castration group ( P <0.05). Conclusion: Androgen replacement therapy can improve the learning and memory ability of mice,which may be achieved by reducing the expression of GFAP and inhibiting the activation of astrocytes.
