摘要 为检测一株杀鲑气单胞菌拮抗菌解淀粉芽孢杆菌X8的安全性,进行了小球藻生长影响试验、急性毒性试验、慢性毒性试验、免疫指标检测、细菌易位试验、溶血试验及抗生素敏感性试验等安全性试验.小球藻生长影响试验采用3个不同浓度的菌液与一定量小球藻共培养72h;急性毒性试验分为高剂量组(109CFU/mL)、中剂量组(108CFU/mL)、低剂量组(107CFU/mL)和生理盐水对照组,一次性注射200μL拮抗菌X8后观察泥鳅在96h内的存活情况;慢性毒性试验分为高剂量组(106CFU/L)、中剂量组(105CFU/L)、低剂量组(104CFU/L)和生理盐水对照组,泥鳅在此浓度的菌液中养殖30d;采用慢性毒性试验中的泥鳅进行免疫指标SOD,ACP的检测.结果表明:小球藻生长影响试验中,淡水小球藻与拮抗菌X8共培养较单独培养时叶绿素a含量降低且差异显著,海水小球藻与拮抗菌X8共培养较单独培养时叶绿素a含量变化差异不显著;急性毒性试验中的泥鳅全部存活,体表及内脏无异常病理变化;慢性毒性试验中的泥鳅均无异常病变,试验组泥鳅体质量较对照组泥鳅明显增长;免疫指标检测中,试验组SOD活性低于对照组活性,且差异达到显著水平(P<0.05),ACP活性高于对照组活性,且差异达到显著水平(P<0.01);拮抗菌X8菌株在泥鳅体内未发生易位和溶血现象;拮抗菌X8对利福平、四环素、头孢克肟、磺胺异亚唑和链霉素不敏感,对诺氟沙星、阿莫西林、青霉素、复方新诺明和庆大霉素敏感.杀鲑气单胞菌拮抗菌X8在泥鳅养殖环境中使用安全,具有良好的应用前景. In order to detect the safety of the antagonistic bacteria X8 against Aeromonas salmonicida , experiments on the effects of Chlorella growth,the acute toxicity test, chronic toxicity test, bacterial translocation test, hemolytic test and antimicrobial susceptibility test were carried out.The growth of Chlorella was influenced by three different concentrations of bacterial solution and a certain amount of Chlorella for 72 hours.Acute toxicity test was divided into higher dose group (10 9 CFU/mL), middle dose group (10 8 CFU/mL), lower dose group (10 7 CFU/mL) and control group (saline solution).After one-time injection of 200 μL, the observation of the loach survival was performed for 96 hours.Chronic toxicity test was divided into higher dose group (10 6 CFU/L), middle dose group (10 5 CFU/L), lower dose group (10 4 CFU/L) and control group (saline solution).Subsequently, the loaches were soaked in the above concentration for 30 days.The immune indexes SOD and ACP of loach in chronic toxicity test were detected.The results showed that in the experiment of Chlorella growth influence, the chlorophyll-a content of freshwater Chlorella and antagonistic bacteria X8 co-culture was lower than that of single culture, and there was no significant difference in chlorophyll-a content between seawater Chlorella and antagonistic bacteria X8 co-culture.All the loach survived in the acute toxicity test, and there were no abnormal pathological changes in the surface and viscera.In the chronic toxicity test, there was no abnormal lesion in the loach.The body weight of the experimental group was significantly higher than that of the control group.The activity of SOD in the experimental group was lower than that in the control group, and the difference was significant ( P <0.05).The activity of ACP in the experimental group was higher than that in the control group, and the difference was significant ( P <0.01).The antagonistic bacteria X8 had no ectopic reaction in loach and no hemolysis occurred.The antagonistic bacteria X8 was
