管花肉苁蓉提取物正丁醇残留量检测及去除工艺优选

摘要 目的:建立管花肉苁蓉提取物正丁醇残留量的检测方法,优选残留正丁醇的最佳去除工艺。方法:以正丁醇残留量为考察指标,运用气相色谱法测定正丁醇残留量,采用正交试验方法,优选最佳的残留正丁醇去除工艺。结果:正丁醇残留量检测方法平均回收率为100.5%,RSD值为1.83%。管花肉苁蓉提取物残留正丁醇最佳去除工艺为每次加水0.8倍,80℃旋转蒸发3次,测得正丁醇残留量符合《中华人民共和国药典》规定限量标准。结论:本正交设计试验所得的正丁醇残留量检测方法准确可靠,优选的正丁醇去除工艺简便、可行,为后续研究提供参考。 Objective: To build a method to detect residual n -butanol and optimize the process for eliminating n -butanol in extract of Cistanche tubulosa . Methods: The residual n -butanol in C.tubulosa extract was detected by GC,and the processing condition for the elimination of n -butanol with residual n -butanol as the index was optimized by using the orthogonal experimental design. Results: The recovery of residual n -butanol was 100.5% with RSD being 1.83%.The optimal process for eliminating n -butanol was adding 0.8-fold water every time and reduced-pressure distillation at 80 ℃ for three times,and the obtained residual n -butanol level was in accordance with the Chinese pharmacopoeia limit. Conclusions: The method for detecting residual n -butanol in C.tubulosa extract is accurate and reliable.It can provide a basis for follow-up study.
