摘要
目的:探究莪术醇对博来霉素诱导的大鼠肺纤维化的干预作用及对TGF-β1和PAI-1表达的影响。方法:45只大鼠随机分为对照组、模型组和治疗组,每组15只,模型组及治疗组给予气管内博来霉素滴注,对照组给予等量生理盐水。造模后隔天治疗组给予5mg/kg莪术醇灌胃给药,对照组、模型组予等量生理盐水灌胃,给药28d后取各组大鼠肺组织进行HE染色和Masson染色检测肺组织纤维化情况;免疫组织化学染色检测TGF-β1和PAI-1表达及分布;检测肺组织羟脯氨酸(HYP)含量。结果:HE染色及Masson染色显示治疗组大鼠肺泡炎和肺纤维化程度均低于模型组,胶原纤维形成少于模型组(P<0.05);模型组大鼠TGF-β1和PAI-1表达量高于对照组;治疗组大鼠TGF-β1和PAI-1表达量均低于模型组(P<0.05);治疗组大鼠肺组织中HYP含量低于模型组(P<0.05)。结论:莪术醇可抑制肺纤维化大鼠肺组织中TGF-β1和PAI-1的表达,缓解博来霉素诱导的大鼠肺纤维化。
Objective:To explore the effect of zedoary on rats with bleomycin-induced pulmonary fibrosis and the expression of TGF-β1 and PAI-1.Methods:The 45 rats were randomly divided into 3 groups,the control group,the model group and the treatment group,with 15 rats in each group.The model group and the treatment group were given bleomycin instillation by trachea.The control group was given equal amount of saline.After modeling,the treatment group was given 5 mg/kg zedoary turmeric for the next day.The other two groups were given equal amount of saline,After 28 days,the lung tissues of each group were taken for HE staining and Masson staining to detect the fibrosis of lung tissue.The expression and distribution of TGF-β1 and PAI-1 were detected by immunohistochemistry.Some lung tissues were used for quantitative determination of hydroxyproline content.Results:HE staining and Masson staining showed that the degree of alveolitis and pulmonary fibrosis in rats after treatment with zedoary turmeric were significantly lower than those in model group,the formation of collagen fibers decreased,and the degree of pulmonary fibrosis decreased significantly(P<0.05).The expression levels of TGF-β1 and PAI-1 in model group increased sharply compared with those in control group.And after 28 days of zedoary,these indexes of the treatment group were significantly decreased(P<0.05).After 28 days of zedoary injection,the HYP content in the lung tissue of the treatment group was significantly lower than that of the model group(P<0.05).Conclusion:Zedoary can inhibits the expression of TGF-β1 and PAI-1 in lung tissue of rats with fibrosis,and it can effectively relieve pulmonary fibrosis induced bleomycin in rats.
