黄芪多糖上调肠道黏膜免疫分子基因表达对口蹄疫特异性抗体生成的促进作用

Acceleration effects of immune molecular genes expression in intestinal mucosa upgrade with Astragalus polysaccharide on specific antibody production of Foot-and-mouth disease

摘要 为进一步探究黄芪多糖对肠道黏膜及口蹄疫疫苗免疫的增强作用,给试验小鼠口服4d黄芪多糖,之后接种口蹄疫商品疫苗,收集二免后1~3周血清,采用间接夹心ELISA法检测特异性免疫球蛋白G(IgG)水平及抗体效价;二免后3周处死小鼠,收集十二指肠组织,用实时荧光定量PCR法检测肠道组织CD80和CD86、主要组织相容性复合体Ⅱ(MHC-Ⅱ)、Toll样受体4(TLR-4)及核因子-κB(NF-κB)p65的mRNA表达水平。结果表明:试验组小鼠口服黄芪多糖二免后第2,3周血清IgG水平及抗体效价显著高于生理盐水对照组(P<0.05);二免后3周,肠道组织CD80、CD86、MHC-Ⅱ、TLR-4及NF-κBp65的mRNA表达水平显著高于生理盐水对照组(P<0.05)。说明口服黄芪多糖能够上调肠道黏膜固有免疫细胞免疫分子基因的表达,从而促进口蹄疫疫苗免疫的抗体反应。 To investigate the effect of Astragalus polysaccharide on immunity of intestinal mucosa and Foot-and-mouth disease vaccine, mice were inoculated with Foot-and-mouth disease vaccine after 4 days orally administrated. The level of serum specific IgG and IgG titer 1-3 weeks after secondary immunization were measured by indirect ELISA. The mice were slaughtered 3 weeks after secondary immunization. The duodenum tissue was collected, the mRNA expression levels of intestinal tissue CD80, CD86, major histocompatibility complexes Ⅱ(MHC-Ⅱ), Toll-like receptor 4(TLR-4) and nuclear factor-κB(NF-κB) p65 were measured by Real time fluorescence quantitative PCR method. The results showed that the level of serum IgG and IgG titer are significantly higher than control group 2,3 weeks after secondary immunization(P<0.05). The level of gene expression of CD80,CD86,MHC-Ⅱ,TLR-4 and NF-κB p65 on 3 weeks past booster immune are also notably higher than control group(P<0.05).It indicates that the gene expression of intestinal mucosal immune molecules of innate immune cells are up-regulated by oral administration Astragalus polysaccharide, thus promot specific antibody immune response of inoculation with Foot-and-mouth disease vaccine.
