摘要
目的探讨Epstein Barr病毒(EBV)编码的microRNA bart7调节鼻咽癌放疗敏感性的机制。方法筛选75例鼻咽癌患者以及同期入院体检的30例健康人群为研究对象。比较两组间鼻咽癌患者EBV/DNA比率。测定鼻咽癌患者的microRNA bart7、EBV VCA IgA、EBV EA IgA、EBV Rta IgG等EBV相关肿瘤标志物阳性状态,分析EBV/DNA与microRNA bart7间的相关性。结果鼻咽癌患者血浆EBV/DNA比率高于健康人群(92.00%vs 3.33%,P<0.05)。放疗后鼻咽癌患者血浆EBV/DNA比率低于放疗前(30.67%vs 92.00%,P<0.05)。放疗后持续缓解的鼻咽癌患者血浆EBV/DNA阳性率低于复发/转移者(12.50%vs 39.22%,P<0.05)。放疗后鼻咽癌患者血浆microRNA bart7、EBV VCA IgA、EBV EA IgA、EBV Rta IgG阳性率均低于放疗前(P<0.05)。放疗后,持续缓解的鼻咽癌患者血浆microRNA bart7、EBV VCA IgA、EBV EA IgA、EBV Rta IgG阳性率均低于复发/转移者(P<0.05)。EBV/DNA阳性率与microRNA bart7阳性率呈正相关(P<0.05)。结论EBV可能通过调节其编码的microRNA bart7水平对鼻咽癌患者放疗敏感性产生影响。
Objective To investigate the mechanism of Epstein Barr virus encoded microRNA bart7 regulating radiosensitivity of nasopharyngeal carcinoma.Methods A total of 75 patients with nasopharyngeal carcinoma and 30 healthy subjects admitted to the hospital were screened.The positive rate of EBV/DNA in patients with nasopharyngeal carcinoma was compared.To investigate the effect of EBV/DNA on the sensitivity of radiotherapy to patients with nasopharyngeal carcinoma,the positive rate of microRNA bart7,EBV VCA IgA,EBV EA IgA,EBV Rta and other related tumor markers were determined.Results The positive rate of plasma EBV/DNA in nasopharyngeal carcinoma patients was higher than that in healthy subjects(92.00%vs 3.33%,P<0.05).The positive rate of plasma EBV/DNA in nasopharyngeal carcinoma patients after radiotherapy was lower than that before radiotherapy(30.67%vs 92.00%,P<0.05).The positive rate of plasma EBV/DNA in patients with nasopharyngeal carcinoma after radiotherapy was deceased(12.50%vs 39.22%,P<0.05).The positive rates of plasma microRNA bart7,EBV VCA IgA,EBV EA IgA and EBV Rta in patients with nasopharyngeal carcinoma after radiotherapy were lower than those before radiotherapy(P<0.05).After radiotherapy,the positive rates of microRNA bart7,EBV VCA IgA,EBV EA IgA and EBV Rta IgG were lower in the response patients than those of the recurrent/metastatic patients.The positive rate of EBV/DNA was positively correlated with the positive rate of microRNA bart7(P<0.05).Conclusion EBV may affect the sensitivity of radiotherapy in patients with nasopharyngeal carcinoma by regulating the microRNA bart7 level.
