摘要
目的探讨蒿本内酯(ligustilide,LIG)早期应用对大鼠脑出血(intracerebral hemorrhage,ICH)的神经保护作用。方法将96只雄性SD大鼠按随机分为被随机分为对照组(Control组)、脑出血+载体组(ICH+Vehicle组)、脑出血+蒿本内酯(ICH+LIG 5mg)处理组和脑出血+蒿本内酯(ICH+LIG 10mg)处理组,每组24只。Control组对右侧基底节区穿刺不注血,另3组均抽取右股动脉自体血100μL注入右基底节区造脑出血模型,药物治疗组术后5min经尾静脉给药,术后每天1次。术后24h,检测脑水含量、检测血脑屏障通透性和电镜观察线粒体的变化。术后72h,HE检测细胞形态和行为学评分。结果术后24h,ICH+Vehicle处理组脑含水量明显高于其它组,ICH+Vehicle处理组EB含量显著高于其它组,药物处理组脑水含量和血脑屏障通透性显著改善,差异均有统计学意义(P<0.05);电镜观察ICH+Vehicle处理组线粒体肿胀程度较其它组明显,药物处理组线粒体肿胀程度显著改善。术后72h,HE染色ICH+Vehicle处理组细胞数明显高于其它组,ICH+Vehicle处理组行为学评分显著低于其它组,药物处理组细胞坏死和行为学评分显著改善,差异均有统计学意义(P<0.05)。结论蒿本内酯早期足量应用能够降低脑出血大鼠脑水肿,改善血脑屏障通透性,减少神经细胞坏死,改善脑出血大鼠神经功能。
Objective To explore the neuroprotective effect of early application of ligustilide on experimental intracerebral hemorrhage (ICH) in rats.Methods 96 male Sprague-Dawley rats were randomly divided into 4 groups: control group,ICH+Vehicle group,ICH+LIG 5 mg intervention group and ICH+LIG 10 mg intervention group,24 in each;except the rats in control group,the rats in the other 3 groups received injection of 100μL of autologous blood extracted from the right femoral artery into the right basal ganglia so as to construct the ICH models,and 5 minutes after surgery,drugs were applied via the tail vein (later once a day);24 hours after surgery,the brain water content and the blood-brain barrier permeability were detected,and the changes of mitochondria were observed by electron microscopy;72 hours after surgery, cell morphology and behavioral scores were detected by HE.Results 24 hours after surgery,the brain water content and the ICH content of the rats in ICH+Vehicle group were higher than those of the rats in other groups;the improvement of the brain water content and blood-brain barrier permeability in drug-treated groups were obvious and the difference was statistically significant (P<0.05);the mitochondrial swelling in ICH+Vehicle group was more severe than that in other groups and it was greatly improved in the drug-treated groups;72 hours after surgery,HE staining indicated that the number of cells was significantly bigger and the behavioral scores were obviously lower in ICH+Vehicle group than those in the other 3 groups,and the nubmber of cells and behavioral scores in drug-treated group were greatly improved (P<0.05).Conclusions Early application of full-dose ligustilide can reduce the cerebral edema in ICH rats,improve the blood-brain barrier permeability,decrease neuronal necrosis and promote the amelioration of nerve function in ICH rats.
