摘要
目的评价肝素接枝对脱细胞血管支架材料结构的影响,及其作为动静脉造瘘移植物的短期抗凝效果。方法采用猪颈动脉作为血管材料来源,通过除垢剂脱细胞法制备脱细胞血管支架材料,对支架材料管腔内层进行层层自组装肝素接枝,并用HE、Masson、EVG、甲苯胺蓝等组织染色和扫描电镜等方法,对支架材料结构及肝素接枝效果进行分析。通过手术开放猪颈部动静脉血管,用半在体动静脉造瘘的实验方法连接血管支架材料,对是否有肝素接枝的不同材料进行3 h的抗凝性能评价。结果层层自组装肝素接枝方法可以在血管支架内层形成均匀的肝素层,对血管脱细胞支架材料的胶原纤维、弹性纤维等基本组织结构无明显影响;脱细胞血管移植物3 h的血栓形成程度,有肝素接枝的材料明显较没有肝素接枝的减轻。结论层层自组装技术肝素接枝后,可以提高脱细胞血管支架材料的短期抗凝效果。
Objective To evaluate the short - term anticoagulant effect of heparin - grafted decellularized vascu- lar scaffold material as a graft of arteriovenous fistula. Methods The porcine carotid arteries were used as the source of vascular material. The decellularized vascular scaffold was prepared by the decellularization method of the detergents. We used layer - by - layer self - assembly technique to graft heparin on the inner lumen of decellularized vascular scaffolds. Histological staining ( HE,Masson,EVG,toluidine blue staining) and scanning electron microscopy were performed to analyze the structure. We exposed the carotid artery and jugular vein of pig by surgery,an in vitro experiment method of arteriovenous fistula was established to observe the anticoagulant performance of both non - heparin - grafted and heparin - grafted decellularized vascular scaffolds for 3 h. Results The layer - by - layer self - assembled heparin grafting method could form a homogeneous heparin layer in the inner layer of vascular scaffolds,without significant damage on the basic tissue structure of collagen fibers and elastic fibers of decellularized vascular scaffolds. The heparin - grafted decellularized vascular scaffolds had significantly less thrombus formation in 3 h than the non - heparin - grafted decellularized vascular scaffolds. Conclusion The layer - by - layer self - assembled heparin grafting technique can improve the short - term an- ticoagulant effect of decellularized vascular scaffolds.
