不同浓度NaClO配合被动超声对根管内细菌生物膜清除效果的研究 被引量:11

Evaluation of different concentration of sodium hypochlorite with passive ultrasonic irrigation against intra-canal biofilm in vitro

摘要 目的:比较被动超声(passive ultrasonic irrigation,PUI)配合不同浓度NaClO杀灭根管内成熟粪肠球菌生物膜的效果。方法:选取人单根管离体牙48颗,标准根管预备后,体外培养14 d粪肠球菌(Enterococcus faecalis)生物膜,并使用不同浓度NaClO(120 s)配合PUI荡洗60 s,即1%NaClO(G1组),1%NaClO+PUI(G2组),2. 5%NaClO(G3组),2. 5%NaClO+PUI(G4组),5. 25%NaClO(G5组),5. 25%NaClO+PUI(G6组)和H2O+PUI(G7组),H_2O(G8组),分别进行细菌计数,及扫描电镜观察评价杀菌效果。结果:细菌计数的差异:G7组和G8组与其它6组差异均有统计学意义(P<0. 05); G5和G6组之间差异无统计学意义(P>0. 05),但是和其它几组具有显著性差异(P<0. 05),G2和G3之间差异无统计学意义(P>0. 05)。扫描电镜观察示PUI能够更有效清除表面细菌生物膜。结论:低浓度NaClO辅助PUI能够有效提高杀菌和清除生物膜效果;高浓度NaClO配合PUI并不能显著提高其杀菌效果,但能更有效地清除成熟粪肠球菌生物膜。 Objective: To evaluate the biofilm eliminating effects of 1%,2. 5% and 5. 25% NaClO with passive ultrasonic irrigation( PUI) against intra-canal biofilm.Methods: All 48 extracted teeth had been selected and constructed 2 weeks Enterococcus faecalis biofilm in vitro,then treated with different concentration of sodium hypochlorite and passive ultrasonic irrigation( PUI),the bactericidal effects had been evaluated,that Group 1 with 1%NaClO,Group 2 with 1%NaClO+PUI,Group3 with 2. 5%NaClO,Group 4 with 2. 5%NaClO+PUI,Group 5 with 5. 25% NaClO,Group 6 with 5. 25%NaClO+PUI,Group 7 with H2 O+PUI and Group 8 with H2O. To further observe the biofilm eliminating effects,the scanning electron microscopy( SEM) was used.Results: There was statistic significantly difference been G7,G8 and other six groups( P<0. 05),no statistic difference between G5 and G6( P>0. 05),either as G2 and G3( P>0. 05).The SEM results indicated that the passive ultrasonic irrigation could help eliminating biofilm. Conclusion: The passive ultrasonic irrigation could significantly contribute to better biofilm eliminating and killing at low concentration of sodium hypochlorite.Although passive ultrasonic irrigation could not enhance the biofilm killing effect,the better root canal eliminating effects had been observed.
