摘要
目的筛选中东亚人群胃癌的易感基因,为胃癌的诊断和治疗提供新思路。方法在基因表达数据库(GEO)中进行检索,利用在线工具GEO2R筛选胃癌的差异表达基因,并对其进行基因本体(GO)富集分析及京都基因与基因组百科全书(KEGG)信号转导通路分析,最后利用STRING数据库对基因相关蛋白相互作用进行关联构建。结果本研究共纳入2个数据集,通过筛选得到40个差异表达基因,其中上调基因34个,下调基因6个,并首次发现ARHGAP11A基因与胃癌相关。通过对差异表达基因的分析发现,涉及的信号通路主要有白细胞转运蛋白迁移、p53信号通路、细胞周期等。结论利用生物信息学方法可有效地筛选出中东亚人群的胃癌易感基因,从而为胃癌发病机制的研究提供依据。
Objective To screen the susceptibility genes of gastric cancer in middle-east Asian population,and providing new ideas for diagnosis and treatment. Methods The gene was searched in the gene expression database (GEO).The differentially expressed genes of gastric cancer were screened by online tool GEO2R,and the GO enrichment analysis and KEGG signal transduction pathway were analyzed.Finally,the gene-related proteins were used in the STRING database. Results On this study,two data sets were included,and 40 differential genes were obtained by screening,including 34 up-regulated genes and 6 down-regulated genes,and the ARHGAP11A gene was firstly found to be associated with gastric cancer.Through analysis of differential genes,it was found that the signaling pathways involved were leukocyte transporter migration,p53 signaling pathway,and cell cycle. Conclusion The susceptibility genes of gastric cancer can be effectively screened out by bioinformatics,which can provide a basis for the study of the pathogenesis of gastric cancer.
