摘要
目的探讨木香油方治疗脑卒中的“成分-靶点-疾病”相关关系。方法从整合药理学平台中检索脑卒中的靶标和木香油方各组成药味的化学成分及其作用靶标,构建“药材-化学成分-靶标-通路”的网络并进行分析。结果检索到木香、胡椒、细辛中共189个活性成分,其药理作用涉及99个核心靶标,其中包括痕量胺相关受体1(TAAR1)、G蛋白α亚基(GNAQ)、人嘌呤能受体(P2RY12)、1型血管紧张素Ⅱ受体基因(ATGR1)、SELE基因、趋化因子受体(CXCR3)、细丝蛋白A(FLNA)、SACS基因等。这些关键靶标可能与神经系统、神经营养因子信号通路、神经退行性疾病、内分泌系统、细胞生长和死亡等相关生物过程和代谢通路有关。结论该研究预测了木香油方治疗脑卒中的关键靶标及参与的相关通路,为进一步研究木香油方治疗脑卒中的作用机制提供了科学依据。
Objective To explore the relationship of “component-target-disease” of Mu-Xiang-You-Fang on treatment of stroke with integrated pharmacological method. Methods Based on the analysis function of integrative pharmacology platform,the compatibility mechanism of this prescription was investigated by constructing the herbs-compounds-targets network. Results A total of 189 active ingredients from Aucklandia, Piper and Asarum were identified. Their pharmacological effects involved 99 core targets, including trace amine-related receptor 1(TAAR1),G protein alpha subunit (GNAQ),human purinergic receptor (P2RY12), angiotensin II receptor type 1(ATGR1),SELE gene,chemokine receptor(CXCR3),filament A (FLNA), SACS gene and so on. These key targets may be related to biological processes and metabolic pathways such as nervous system,neurotrophic factor signaling pathway,neurodegenerative diseases,endocrine system,cell growth and death. Conclusion This study predicted the key targets and related pathways of Mu-Xiang-You-Fang in the treatment of stroke,and provided a scientific basis for further study of the mechanism of Mu-Xiang-You-Fang in the treatment of stroke.
