摘要
目的建立活体犬上颈椎融合模型并观察术后3个月C2-3内I型胶原蛋白的变化。方法将成年雄性健康家犬12只随机分为实验组和对照组(各6只),实验组行寰枢椎融合术,对照组不做处理。术后3个月处死动物并获取C2-3椎间盘,行HE染色观察椎间盘纤维环的形态,并进行评分。行椎间盘免疫荧光染色观察I型胶原蛋白。采用Image-Pro Plus 6.0软件将荧光图像转换为灰度图像,并进行光密度测量,获得每个椎间盘的平均光密度值。对上述两组椎间盘进行独立样本t检验。结果实验组C2-3椎间盘纤维环排列散乱,对照组C2-3椎间盘纤维环排列整齐,两组椎间盘平均评分实验组为1.5,对照组为0.5,两组评分比较差异有统计学意义(χ^2=3.105,P<0.05)。实验组内I型胶原蛋白的光密度值为(391 052.98±18 869.81),较对照组的光密度值(312 945.42±22 400.47)显著升高(P<0.001)。结论上颈椎融合能加速C2-3椎间盘的退变.
Objective To establish an in vivo canine upper cervical spine fusion model and to evaluate the expression level of type I collagen in C2-3 disc after upper cervical spine fusion. Methods Twelve healthy adult male canines were included and randomly divided into experimental or control group, with 6 canines in each group. Upper cervical spine fusion was conducted on the canines in the experimental group while leaving the control group untreated. Canines were euthanatized at the third month after the operation. HE and immunofluorescence staining for type I collagen in C2-3 disc were performed. Two independent samples t test was used to compare the optical density of the C2-3 disc in two groups. Results Disordered annulus fibrosus of the experimental group while orderly annulus fibrosus of the control group were observed. Significant increased score was observed in experiment group comparing with the control group. The average score of the intervertebral disc in the experimental group was 1.5 while the control group was 0.5. The difference between the score in two groups was statistically significant (χ^2=3.105, P<0.05). A significant increased optical density in the experimental group (391 052.98±18 869.81) was observed compared with the control group (312 945.42±22 400.47)(t=6.532, P<0.001). Conclusion Upper cervical spine fusion could accelerate the degeneration of the C2-3 disc.
