多房棘球绦虫钙网蛋白结构和功能的生物信息学研究 被引量:3

摘要 为进一步研究多房棘球绦虫的致病性、药物靶点以及疫苗诊断试剂的开发提供理论依据,应用生物信息学方法预测分析多房棘球绦虫(Echinococcus multilocularis,Em)钙网蛋白(Calreticulin,CRT)的结构和功能。通过基因组数据库收集数据确定Em-CRT基因和氨基酸序列;利用生物信息学软件分别对Em-CRT进行蛋白质基本性质、翻译后修饰位点、功能域、亚细胞定位、二/三级结构、亲/疏水性、抗原表位以及进化关系等进行预测和分析。结果表明:Em-CRT基因转录本长度为1 188 bp,蛋白全长为395个氨基酸残基,分子量大小约为45.44 kDa,等电点(pI)为4.47,为稳定蛋白,亲水性较高,可溶于水;具有信号肽和信号肽酶剪切位点,为膜外蛋白;Em-CRT具有N端糖基化位点,酪氨酸激酶(TK)Ⅱ磷酸化位点,cAMP和cGMP依赖蛋白激酶磷酸化位点,N端肉豆蔻酰化位点,蛋白激酶C磷酸化位点,酪氨酸激酶磷酸化位点;二级结构以无规则卷曲和α螺旋为主;Em-CRT具有良好的T,B细胞抗原表位。所以,Em-CRT对多房棘球绦虫个体发育具有重要的作用,且与多房棘球蚴感染免疫密切相关,可参与感染过程或宿主免疫应答,具有作为诊断试剂抗原分子的价值和潜在的疫苗候选分子与新的药物靶标的应用前景。 The structure and function of calreticulin(CRT) of Echinococcus multilocularis were analyzed by bioinformatics method,which provided theoretical basis for further study on the pathogenicity,drug targets,vaccines and diagnostic reagents.The Em-CRT gene and amino acid sequence were determined by collecting data from the genomic database.The prediction and analysis of the basic properties,post-translational modification,functional domains,subcellular localization,secondary and tertiary structure,pro-/hydrophobicity,antigenic epitopes and evolutionary relationships of Em-CRT were performed by using various bioinformatics software.The results showed that the transcript of Em-CRT gene was 1 188 bp and 395 amino acid residues;the molecular weight was about 45.44 kDa,and the isoelectric point(pI) was 4.47,which was stable and high hydrophilicity protein and soluble in water.The Em-CRT protein was with signal peptide and signal peptidase cleavage sites,and was an extramembrane protein.The Em-CRT also had N-glycosylation site,cyclic Adenosine monophosphate(cAMP)-and cyclic guanosine monophosphate(cGMP)-dependent protein kinase phosphorylation site,tyrosine kinase(TK) II phosphorylation site,N-myristoylation site,protein kinase C phosphorylation site and tyrosine kinase phosphorylation site.The secondary structure of Em-CRT was random coil and alpha helix,and had epitopes of T,B cell.Em-CRT plays an important role in the development of E.multilocularis,and is closely related to the infection of E.multilocularis infection,may participate in the infection process and host immune response.Em-CRT could be used as a diagnostic antigen molecule,and potential vaccine candidate and new drug target.
