摘要
气溶素和溶血素是β-溶血性嗜水气单胞菌的重要毒力因子。为建立检测食用鱼类产品中β-溶血性嗜水气单胞菌气溶素基因(aerA)和溶血素基因(ahh1)的技术,本文采用多重PCR技术检测28个来源于食用鱼类产品的β-溶血性嗜水气单胞菌样本(包括鲤鱼片、水、鲫鱼片、鳟鱼片、鲢鱼片、鲑鱼片和带鱼片),并用标准菌株嗜水气单胞菌ATCC7965和分离于死蟒蛇的带有β-溶血性毒素基因的嗜水气单胞菌作为对照。结果表明,所有菌株样本(100%)被证实携带嗜水气单胞菌16SrRNA基因(356个碱基对),所有嗜水气单胞菌样本(100%)被证实携带ahh1毒性基因(130个碱基对),26个嗜水气单胞菌样本(93%)被证实携带aerA基因(309个碱基对)。本研究证实β-溶血性嗜水气单胞菌菌株广泛存在于供人类食用的鱼类产品中,对消费者产生了一定的健康风险。
Aerolysin and haemolysin are viewed as important virulence factors related to the entero-toxigenicity of β-haemolytic Aeromonas hydrophila. In order to establish a method for detection of genes coding aerolysin(aerA)and haemolysin(ahh1)in β-haemolytic A. hydrophila strains isolated from edible fish products,a multiplex PCR was applied to detect the β-haemolytic A. hydrophila strains isolated from 28 samples of edible fish products(including carp cutlets,water,crucian cutlets,trout cutlets,silver carp cutlets,trout cutlets and hailtail cutlets),and the study used a reference A. hydrophila strain(ATCC 7965)and an A. hydrophila strain isolated from a dead anaconda,as positive controls with strong β-haemolytic activity. The results showed that the chromosome gene 16S rRNA(356 bp)was confirmed in all A. hydrophila strains(100%),the gene ahh1(130 bp)was confirmed in all A. hydrophila strains(100%),and the gene aerA(309 bp)was confirmed in 26 A. hydrophila strains(93%). The presence of β-haemolytic A. hydrophila strains in edible fish products cuases a risk to the consumer′s health.
