知母不同药用部位体外抗肿瘤、抗炎及抗氧化的作用研究 被引量:17

Research of Different Medicinal Parts of Anemarrhena Asphodeloides on Anti-tumor,Anti-inflammatory and Anti-oxidation in vitro Based on Pharmacodynamics

摘要 目的:比较知母药材不同药用部位体外抗肿瘤、抗炎及抗氧化的作用,为知母进一步的合理应用和开发提供参考。方法:采用HPLC色谱法建立知母药材不同药用部位的指纹图谱,并进行相似度评价;采用MTT法研究知母不同药用部位对人胃癌HGC-27细胞、人肝Hepg-2细胞和人结肠癌Caco-2细胞体外增值抑制作用的影响;采用LPS诱导Raw 264.7细胞炎症模型,MTT法测定细胞活力,Griess试剂盒测定细胞分泌一氧化氮(NO)的水平,探讨知母不同药用部位的体外抗炎活性;采用二苯基苦酰肼基(DPPH)、2,2′-连氨-(3-乙基苯并噻唑啉-6-磺酸)二氨盐(ABTS)自由基清除法观察知母不同药用部位的体外抗氧化作用。结果:HPLC指纹图谱显示,知母4个不同药用部位根、茎、叶、花的化学成分具有一定的差异性;体外药效结果显示,知母不同药用部位作用于不同肿瘤细胞后均呈现不同程度的增值抑制作用,具有一定的差异性;知母不同药用部位均能促进RAW264.7巨噬细胞的增殖,不同药用部位对LPS诱导的巨噬细胞分泌一氧化氮均有不同程度的抑制作用;知母花对DPPH及ABST的清除能力最强,其它部位活性顺序依次为知母叶、茎、根。结论:知母根、叶、花为抗肿瘤有效药用部位;知母不同药用部位均具有显著的抗炎、抗氧化作用,以知母茎抗炎作用最强,知母花抗氧化作用最强;本研究为知母不同药用部位针对不同疾病的合理使用提供实验依据和实验方法。 Objective: To provide references for further rational application and development of Anemarrhena Asphodeloides by comparing the anti-tumor, anti-inflammation and anti-oxidant effects of different medicinal parts of Anemarrhena Asphodeloides in vitro. Methods: HPLC chromatography was used to establish the fingerprint of different medicinal parts of Anemarrhena Asphodeloides and similarity evaluation was conducted. MTT method was used to study the effect of different medicinal parts of Anemarrhena Asphodeloides on proliferation inhibition in vitro of human gastric cancer hgc-27 cells, human liver hepg-2 cells and human colon cancer caco-2 cells. LPS was used to induce Raw 264.7 cell inflammation model, and cell viability was detected by MTT assay. Levels of nitric oxide(NO) secreted by cells were determined by Griess kit to explore the anti-inflammatory activity of different medicinal parts of Anemarrhena Asphodeloides in vitro. The anti-oxidant effect of different medicinal parts of Anemarrhena Asphodeloideswas was observed by using DPPH, ABST free radical scavenging method in vitro. Results: HPLC fingerprint showed that the chemical components of the roots, stems, leaves and flowers of the Anemarrhena Asphodeloides were different from each other. In vitro, the pharmacodynamic results showed that different parts of Anemarrhena Asphodeloides showed different degree of value-added inhibition after acting on different tumor cells, and there were some differences. The proliferation of RAW264.7 macrophages can be promoted in different medicinal parts of Anemarrhena Asphodeloides, which can inhibit the secretion of NO by LPS. The removal ability of DPPH and ABST was the strongest in the flowers of Anemarrhena Asphodeloides, and the activity in other parts was successively followed by leaves, stem and root of Anemarrhena Asphodeloides. Conclusion: The rhizome, leaf and flower of Anemarrhena Asphodeloides are effective parts for anti-tumor medicine. The Anemarrhena Asphodeloides has significant anti-inflammatory and anti-
