沉默miR-345联合5-FU对胃癌MGC803细胞增殖能力及凋亡的影响

摘要 目的 观察沉默miR-345联合5-FU对胃癌MGC803细胞增殖及凋亡的影响。方法采用脂质体介导转染miR-345反义寡核苷酸至胃癌细胞MGC803以沉默miR-345表达。应用40μg/ml5-FU单独处理胃癌细胞MGC803或联合miR-345沉默组,克隆形成实验观察细胞的增殖能力,流式细胞仪检测细胞凋亡水平。结果脂质体介导转染miR-345反义寡核苷酸至胃癌细胞MGC803可以抑制miR-345的表达;细胞克隆形成实验显示,反义miR-345+5-FU联合组与单独5-FU处理组相比,克隆形成率[(4.29±0.83)%vs(7.68±0.34)%]显著降低(P<0.01);流式细胞凋亡检测表明,反义miR-345+5-FU联合组细胞凋亡率显著高于反义miR-345组、5-FU组(P<0.01)。结论沉默miR-345联合5-FU能抑制5-FU对MGC803细胞增殖能力,并促进其凋亡。 Objective To observe the effect of miR-345 silence combined with 5-FU on proliferation and apoptosis of gastric cancer cell MGC803 in vitro. Methods miR-345 antisense oligonucleotides were transfected into MGC803 cells. Gastric cancer MGC803 cells were treated with 40 μg/ml 5-FU or combined with miR-345 silence. The proliferation capacity was detected by the cell cloning and the percentage of apoptotic cell was measured by flow cytometry. Results The expression of miR-345 in MGC803 cells transfected antisense miR-345 was significantly reduced. The experiments showed that miR-345 silence combined with 5-FU group had lower percentage of cell cloning than with 5-FU alone group [(4.29±0.83)% vs(7.68±0.34)%](P<0.01), and had higher cell apoptotic rate than miR-345 silence group or 5-FU alone group(P<0.01). Conclusion miR-345 silence combined with 5-FU has a better effect on inhibiting proliferation and promotes the apoptotic rate of MGC803 cell than 5-FU.
