摘要
目前食用菌的交配型鉴定通常采用单核体两两交配后镜检观察锁状联合的方式进行,工作量大,耗时长且易产生人为误检。本研究以香菇(Lentinula edodes)菌株"申香215"为例,通过对需鉴定交配型的单核体的双核体亲本菌株进行基因组重测序,获得双核体交配型A和B因子区域的序列比对信息,对交配型因子等位基因内部的SNP位点进行统计分析,选择合适位点设计引物,采用等位基因特异PCR(Allele-specificPCR,AS-PCR)技术,根据扩增结果可快速确定单核体的交配型并排除双核体和杂合体。用该方法鉴定香菇单核体的交配型,提高了鉴定效率和准确率,该方法可作为分子标记辅助育种的一种手段,为食用菌遗传育种工作提供新的技术支撑。
Currently, the mating type of edible fungi was usually identified by microscopy examination of clamp-connection after mon-mon crossing, which is labor-intensive, time-consuming and prone to artificial detection errors. Taking Lentinula edodes strain "Shenxiang 215" as an example, alignment information of factor A and factor B regions of dikaryon mating type was obtained by genome re-sequencing of the monokaryon and dikaryon parent strains whose mating type requiring identification. SNP loci in mating factors alleles were statistically analyzed, and suitable loci were selected to design primers. Allele-specific PCR (AS-PCR) was used to determine the mating type of monokaryons and to exclude the dikaryons and heterozygotes. The proposed method improved the efficiency and accuracy of identification of the mating type of Lentinus edodes monokaryons. This method may serve as a molecular marker-assisted breeding technique and provide new technical support for the genetic breeding of edible fungi.
