白术配伍桑白皮对肾病综合征大鼠尿液代谢组学的影响 被引量:12

摘要 目的:通过对阿霉素诱导的肾病综合征大鼠尿液代谢物谱的检测,探讨白术配伍桑白皮对肾病综合征大鼠体内代谢的干预作用。方法:采用超高效液相-四级杆串联飞行时间质谱(UHPLC-QTOF-MS)技术分析正常对照组、模型组、贝那普利组、白术组、桑白皮组及白术配伍桑白皮组大鼠的尿液样本。通过SIMCA-P 13.0软件进行主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA),研究白术配伍桑白皮后对肾病综合征大鼠体内代谢的影响并确定与之相关的潜在生物标志物,构建代谢网络。结果:与模型组比较,给药后大鼠尿液代谢轮廓趋向正常对照组,32个与肾病综合征相关的差异标志物水平有向正常水平回调的趋势,且白术配伍桑白皮治疗相比于仅给白术或桑白皮有更为明显的回调趋势。受干扰的代谢通路主要涉及色氨酸代谢、精氨酸和脯氨酸代谢、牛磺酸和亚牛磺酸代谢、2-氧代羧酸代谢等。结论:白术与桑白皮配伍应用后,在一定程度上可改善肾病综合征模型大鼠体内的代谢紊乱,且相比于单独使用白术或桑白皮有更好的疗效。 Objective:To explore the role of metabolic intervention in nephrotic syndrome rats after administrating Atractylodes macrocephala combining with Morus alba through the detection of doxorubicin-induced nephrotic syndrome rat urine metabolites profile.Methods:The urine samples of normal group,model group,benazepril group,Atractylodes macrocephala group,Morus alba group and Atractylodes macrocephala combining with Morus alba group were analyzed by ultrahigh performance liquid chromatography-quadrupole tandem time of flight mass spectrometry(UHPLC-QTOF-MS).Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were performed by SIMCA-P13.0 software.The effect of Atractylodes macrocephala combining with Morus alba on the metabolism of nephrotic syndrome rats was studied and identified the potential biomarkers associated with them and metabolic network was established.Results:Compared with the model group,the metabolic profile of the urine tended to the normal group after administration,32 differential markers associated with nephrotic syndrome tended to rebound to their normal levels,and compared to Atractylodes macrocephala group or Morus alba group,the administration of Atractylodes macrocephala combining with Morus alba had a more pronounced callback trend.The metabolism of tryptophan,arginine and proline metabolism,taurine and hypotaurine metabolism,2-oxocarboxylic acid metabolism and other metabolic pathways were significantly disrupted.Conclusion:Atractylodes macrocephala combining with Morus alba can improve the metabolic disorders in rats with nephrotic syndrome model,and has a better therapeutic effect compared with Atractylodes macrocephala or Morus alba alone.
