摘要
探究毛菊苣不同生长期不同药用部位活性成分秦皮乙素的含量,为确定其最合适的采收季节提供理论依据.本研究于5、6、7、8月采集毛菊苣根、茎、叶等样品,采用高效液相色谱法测定秦皮乙素含量.色谱条件:色谱柱为watersXbridgeC18(4.6mm×250mm,4μm);柱温为25℃;流动相为甲醇-0.2%甲酸(15∶85);流速为1mL/min,检测波长为345nm.研究结果表明,毛菊苣根、茎、叶等不同部位秦皮乙素含量存在极显著性差异(p=0.000),其中茎中含量均较高,其次是根和叶;不同生长期毛菊苣根、茎和叶中秦皮乙素的含量也存在显著性差异(p=0.047),其中6月达到最高值(分别为16.188、24.149和10.455μg·g^-1),含量符合以下规律:5月<8月<7月<6月.毛菊苣不同药用部位不同生长期秦皮乙素含量随着生长发育进程而发生变化.考虑毛菊苣全草入药时以秦皮乙素含量为指标的话,以6~7月枝叶茂盛时采收,有效成分含量最高.
To study contents of aesculetin in different medicinal parts and growth stage of Cichorum glandulosum Boiss.et Huet and to provide a theoretical basis for confirming an optimal recovery season,we collected the root,stem and leaf samples from May to August for this study,High performance liquid chromatography (HPLC) method was used to determined aesculetin contents.The waters Xbridge C18 (4.6 mm×250 mm,4 μm) was used;the column temperature was set at 25 ℃;the detection wavelength was 345 nm;the mobile phase consisted of methanol-0.2% formic acid,and the flow rate was 1 mL/min.The results showed that there were significant differences in the content of aesculetin in roots,stems and leaves of Cichorum glandulosum Boiss.et Huet ( p =0.000),in which the aesculetin was the highest in stems,then the roots and leaves.The content of aesculetin in roots,stems and leaves of Cichorum glandulosum Boiss.et Huetat at different growth stages was also significantly different and reached the highest level in June (16.188,24.149 and 10.455 μg·g ^-1 respectively).The content conforms to the following rules:May<August<July<June.The content of aesculetin in different medicinal parts of Cichorum glandulosum Boiss.et Huet varied with the growth processes.Considering the content of aesculetin as an index when the whole herb of Cichorum glandulosum Boiss.et Huet is used as medicine,the effective component content is the highest when the branches and leaves are harvested from June to July.
