摘要
目的:研究白露汤含药血清对人肺癌A549细胞活力、凋亡及生长周期的影响。方法:将人肺癌A549细胞培养至对数生长期,分别加入白露汤低、中、高剂量含药血清,空白对照组加入含10%胎牛血清的RPMI1640培养液,采用CCK-8法检测细胞活力;Western-blot法测定人肺腺癌A549凋亡细胞Bax、Bcl-2、C-caspase、HIF-1α及β-actin蛋白的表达;流式细胞术检测细胞周期。结果:与空白对照组比较,白露汤各给药组的A549细胞活力在24、48、72 h均明显降低(P<0.05),且呈剂量依赖性。作用24、48、72 h后,白露汤各给药组均出现不同程度的细胞凋亡。流式细胞术检测显示,白露汤作用后,可使人肺腺癌A549细胞其发生早期凋亡(P<0.05)。结论:白露汤可抑制人肺癌A549细胞活力、诱导细胞凋亡、延缓细胞周期进程,这可能与下调GLUT1、HK2、PKM2、LDH-A、HIF-1α和β-actin等表达有关。
Objective: To study the effect of serum containing Bailu Decoction on the activity, apoptosis and cell cycle of human lung cancer A549 cells. Methods: Human lung cancer A549 cells were cultured to logarithmic phase. The cells were added into serum containing Bailu Decoction of low-dose, middle-dose and high-dose respectively, and the blank group was added with RPMI 1640 medium containing 10% fetal bovine serum. The cell viability was detected by CCK-8 method;the expressions of Bax, Bcl-2, C-caspase, HIF-1α and β-actin protein in human lung adenocarcinoma A549 cells were detected by Western-blot method;the cell cycle was detected by flow cytometry technique. Results: Compared to the blank control group, the A549 cell activities of each Bailu Decoction group were significantly reduced at 24 h, 48 h, and 72 h ( P <0.05), which showed a dose-dependent relation. After 24 h, 48 h, and 72 h of the medication, cell apoptosis was found in all Bailu Decoction groups. Flow cytometry results showed that the early apoptosis of A549 cells in human lung adenocarcinoma could be induced by Bailu Decoction( P <0.05). Conclusion: Bailu Decoction can inhibit the activity of A549 cells, induce apoptosis, and delay the process of cell cycle, which may be related to down-regulating the expressions of GLUT1, HK2, PKM2, LDH-A, HIF-1α and β-actin.
