摘要
目的探究F框/WD-40域蛋白7(F-Box and WD40 domain protein 7,Fbxw7)与异柠檬酸脱氢酶1 ( isocitrate dehydrogenase 1,IDH1)在胶质瘤组织中的表达情况。方法用免疫组织化学法,蛋白质印迹法观察86例胶质瘤患者的胶质瘤组织和30例头颅外伤去骨瓣内减压患者(对照组)脑组织中Fbxw7、IDH1的表达与分布,其中低级别胶质瘤48例(WHOⅠ-Ⅱ)、高级别胶质瘤(WHO Ⅲ-Ⅳ)38例,分析免疫组化、蛋白质印迹中Fbxw7、IDH1的表达的差异,分析Fbxw7的表达与可能的临床病理参数(年龄、性别、肿瘤直径、病理分级、头晕病史、癫痫情况、术前KPS评分、胶质瘤细胞成分、IDH1蛋白表达、肿瘤部位)之间的关系。结果胶质瘤组织中Fbxw7的表达明显高于对照组(t=-13.251, P<0.01),且Fbxw7在胶质瘤组织中随着病理级别的升高而表达降低(χ2=8.017, P<0.01);胶质瘤组织中IDH1的表达高于对照组(t=-10.838, P<0.01), IDH1在胶质瘤组织中随着病理级别的升高而升高(χ2=4.282, P=0.036);Fbxw7的表达与胶质瘤直径,病理级别、癫痫发作、IDH1表达有关(χ2=10.994、8.017、14.918、30.454,P<0.05)。结论 Fbxw7蛋白在不同级别胶质瘤组织表达的差异,可能为了解胶质瘤分子学机制提供新思路。
Objective To investigate the expression of F-box and WD40 domain protein 7 (Fbxw7) and isocitrate dehydrogenase 1 (IDH1) in glioma tissues. Methods The expression and distribution of Fbxw7 and IDH1 in 86 glioma patients including 48 cases of low grade glioma (WHO I-II) and 38 cases of high grade glioma (WHO III-IV) and 30 craniotomy patients were included in the present study. Immunohistochemistry and Western blotting was used to analyze the different expression levels of Fbxw7 and IDH1 in immunohistochemistry and protein imprinting. A correlation analysis was conducted to examine the relationship between Fbxw7 expression and pathological parameters (age, sex, tumor diameter, pathological grade, dizziness history, epilepsy, preoperative KPS score, glioma cell composition, IDH1 protein expression and tumor location). Results The expression of Fbxw7 was significantly higher in glioma tissues than in control tissues (t=-13.251, P<0.01). The expression of Fbxw7 decreased with the increase of pathological grade (χ2=8.017, P<0.01). The expression of IDH1 was higher in glioma tissues than in control tissues (t=-10.838, P<0.01). The expression of IDH1 in glioma tissue increased with the increase of pathological grade (χ2=4.282, P=0.036). The expression of Fbxw7 was correlated with the diameter of glioma, pathological grade, seizure and expression of IDH1 (χ2= 10.994, 8.017, 14.918, 30.454, P<0.05). Conclusion The differential expression of Fbxw7 protein in different grade glioma tissues may provide a new clue for the molecular mechanism of glioma.
