小麦内生耐盐菌株YN1的分离鉴定及其生物学特性研究 被引量:2

摘要 本试验从山东无棣县盐碱地健康小麦茎基部分离到1株内生细菌YN1,对其进行菌落形态观察和16SrDNA分子鉴定,并分析其固氮能力、产铁载体能力和产IAA能力,测定了150mmol/LNaCl胁迫下经菌株处理后小麦的耐盐能力。结果表明,该菌株为成团泛菌(Pantoea agglomerans),具有固氮和产铁载体能力,其在0、100mmol/LNaCl条件下IAA产量分别为112.06、153.81μg/mL。与CKI相比,YN1处理的小麦幼苗在150mmol/LNaCl胁迫下株高、根长等显著增加,叶绿素、类胡萝卜素、脯氨酸含量分别提高77.23%、78.55%、47.90%,CAT、POD和SOD活性分别提高74.7%、27.1%和64.2%,而丙二醛含量下降35.3%,表明YN1处理可提高小麦的耐盐能力。 YN1, an endophytic bacterium strain, was isolated from basal stem of healthy wheat in Wudi saline-alkali soil of Shandong Province. The strain was identified by observation of colony morphology and molecular identification of 16 S rDNA,and its nitrogen fixation capacity, siderophore-producing capacity and IAA production capacity were analyzed. The salt tolerance of wheat treated by YN1,strain was tested under the stress of 150 mmol/L NaCl. The results showed that the strain was Pantoea agglomerans and had the ability of nitrogen fixation and siderophore production. The output of IAA was 112.06 μg/mL and 153.81 μg/mL at 0 mmol/L and 100 mmol/L NaCl(W/V), respectively. After treated with YN1, the plant height and root length of wheat seedlings significantly increased compared with the CK1 at 150 mmol/L NaCl stress. Also, the content of chlorophyll, carotenoid and proline increased by 77.23%, 78.55% and 47.90%, respectively. The enzyme activity of CAT, POD and SOD increased by 74.7%, 27.1% and 64.2%, respectively, however, the content of MDA decreased by 35.3%. The results showed that YN1 treatment could improve the salt tolerance of wheat.
