蓝莓鲨烯合酶基因的克隆与表达分析

摘要 鲨烯合酶基因(SQS)在植物三萜类化合物合成途径中起着重要作用,是上游代谢通路中的关键基因。本研究采用RT-PCR技术从‘喜来’蓝莓根中克隆得到VcSQS基因,序列全长1489bp,序列分析结果表明,其含有1242bp的开放阅读框,编码413个氨基酸。氨基酸同源序列分析表明,蓝莓与其它物种SQS蛋白氨基酸序列相似性很高,与山茶科的茶相似性最高,达90.58%,与葡萄相似性达87.92%。荧光定量试验结果表明,SQS基因在蓝莓叶中的表达量最高,芽中的最低。 The squalene synthase gene (SQS) plays an important role in the synthesis of plant triterpenoids and is a key gene in the upstream metabolic pathway. In this study, VcSQS gene was cloned from the root of ‘Xilai’ blueberry by RT-PCR. The full length of VcSQS gene was 1 489 bp. Sequence analysis showed that it contained an open reading frame of 1 242 bp, encoding 413 amino acids. Amino acid homologous sequence analysis showed that the amino acid sequence similarity of SQS protein between blueberry and other species was very high, and its similarity with Camellia sinensis was the highest, up to 90.58%, and the similarity with Vtis vinifera was 87.92%. Fluorescence quantification experiments showed that the SQS gene had the highest expression in blueberry leaves and the lowest expression in buds.
